摘要
目的:运用网络药理学方法探究寒喘祖帕颗粒治疗哮喘的作用机制。方法:利用TCMSP数据库和中国知网检索寒喘祖帕颗粒中9个药物的化学成分,利用TCMSP数据库的靶点预测功能搜集9个药物所含化学成分的潜在作用靶点,运用Cytoscape 3.7.2构建寒喘祖帕颗粒的药物-成分-靶点-疾病网络图,并对潜在的疾病靶点进行蛋白相互作用分析,运用Bioconductor生物信息软件包对潜在疾病靶点进行GO富集分析与KEGG通路富集分析,探究寒喘祖帕颗粒治疗哮喘的核心作用靶点、生物过程与信号通路。结果:药物-成分-靶点-疾病网络图包含9个药物,300个成分,374个靶点;GO分析共富集到855条结果,其中783条富集在生物过程,16条富集在细胞组分,56条富集在分子功能;KEGG通路分析共富集到110条结果。结论:寒喘祖帕颗粒可以通过IL-4、RAF1等基因作用于T细胞受体信号通路、JAK-STAT信号通路和Fc-epsilon-RI信号通路,从而发挥治疗哮喘的作用。
Objective:To explore the mechanism of Hanchuan Zupa granule in the treatment of asthma.Methods:Tcmsp database and CNKI were used to retrieve the chemical components of 9 drugs in Hanchuan Zupa granules.The potential targets of chemical components in 9 drugs were collected by the target prediction function of tcmsp database.The drug component target disease network map of Hanchuan Zupa granules was constructed by using Cytoscape 3.7.2,and the potential disease targets were analyzed by protein interaction,Go enrichment analysis and KEGG pathway enrichment analysis of potential disease targets were carried out by Bioconductor bioinformatics software package to explore the core target,biological process and signal pathway of Hanchuan Zupa granule in the treatment of asthma.Results:The network of herb-component-target-disease included 9 herbs,300 ingredients and 374 targets.There were 855 results in GO analysis,including 783 in biological process,16 in cell component and 56 in molecular function;KEGG pathway analysis enriched 110 results.Conclusion:Hanchuan Zupa granule can act on T cell receptor signaling pathway,JAK-STAT signaling pathway and Fc epsilon RI signaling pathway through IL-4,raf1 and other genes,so as to play a role in the treatment of asthma.
