基于大鼠离体心脏的附子组分强心作用初探 被引量:11

摘要 目的比较附子不同组分(附子总碱、附子水溶性碱和附子精多糖)对离体大鼠衰竭心脏的影响。方法使用低钙K-H液灌流制备离体大鼠衰竭心脏模型,然后分别给予附子不同组分进行干预,并记录分析心率(HR)等血流动力学指标;并用ELISA法测定灌流液中CK-MB、cTn-T水平和Ca2+含量。结果与给药前比较,附子不同组分对离体大鼠衰竭心脏的LVSP、SD、DD的恢复率显著增大(P<0.05),而三个组分均可使衰竭心脏的HR显著减慢(P<0.05);灌流液中CK-MB、cTn-T水平和Ca2+含量均显著下降(P<0.01)。结论2 g/L附子水溶性碱、附子总碱和附子精多糖都对低钙引起的离体大鼠衰竭心脏有强心作用,可改善心功能和减轻心肌损伤。 Objective To compare the effects of different components of aconite(total aconite base,aconite water-soluble base and aconite polysaccharide)on isolated rat heart failure.Methods An isolated rat heart model was prepared by perfusion with low-calcium KH solution,and then the aconite-soluble base,total aconite base,and aconite polysaccharide were used to intervene the isolated heart.The physiological recorder was used.Record heart rate(HR),left ventricular systolic blood pressure(LVSP),systolic duration(SD),diastolic duration(DD),maximum left ventricular pressure rise/fall rate(+dp/dtmax,-dp/dtmax),calculate and analyze the heart failure rate and recovery rate before and after administration;and measure the levels of CK-MB,cTn-T and Ca2+in perfusate by enzyme-linked immunoassay(ELISA).Results The heart of isolated rats perfused with low-calcium KH solution showed accelerated heart rhythm,significantly decreased LVSP and±dp/dtmax(P<0.05),that is,decreased cardiac function;compared with pre-administration,aconite was soluble in water at the same concentration.The recovery rates of alkaloids,aconite total bases,and aconite extract polysaccharides on LVSP and SD of isolated rat heart failure were significantly increased(P<0.01).Aconite water soluble alkali and aconite extract polysaccharides exhibited extremely high recovery rates for DD.Significantly increased(P<0.01),and all three components can slow the heart rate of isolated hearts,which is not significantly different from that of normal hearts,but has no statistical significance with the recovery rate of failed hearts.The levels of CK-MB and cTn-T in perfusion fluid were significantly increased(P<0.01),and the content of Ca2+had no significant change.After different components of aconite were administered,the levels of CK-MB,cTn-T and Ca2+in perfusion fluid were all Significantly decreased(P<0.01).Conclusion 2g/L aconite water-soluble base,total aconite base and aconite polysaccharide all have cardiotonic effect on isolated heart caused by hypocalcemia in rats,wh
