盐酸罗哌卡因纳米结构脂质载体的制备和质量评价 被引量:1

Preparation and quality evaluation of ropivacaine-loaded nanostructured lipid carriers

摘要 目的制备包载盐酸罗哌卡因的纳米结构脂质载体(RP-NLC)并对其进行质量评价。方法采用熔融-超声法制备RP-NLC,通过正交实验优化RP-NLC处方,并对优化后的处方进行质量评价。结果 RP-NLC外观呈类球形,粒径(117.36±12.27)nm,多聚分散度(PDI)为0.275±0.032,粒径分布均一,包封率为(66.28±1.29)%,体外释药具有缓释特征,在4℃条件下放置3个月,粒径、PDI和包封率无明显变化。结论所制备的RP-NLC具有较好的理化性质和缓释作用。 Objective To prepare ropivacaine-loaded nanostructured lipid carriers(RP-NLCs)and evaluate their quality. Methods RP-NLCs were prepared by the melt-ultrasound method,and the formulation was optimized with orthogonal design. Furthermore,the quality of the optimal formulation was evaluated. Results The average particle size of RP-NLCs was(117.36±12.27)nm,polydispersity index(PDI)was 0.275±0.032,and the ropivacaine entrapment efficiency was(66.28±1.29)%. The profiles of RP-NLCs in vitro release had the features of sustained release. The particle size,PDI and zeta potential were also stable during 3 months of incubation at 4℃. Conclusion The prepared RP-NLCs have good physicochemical properties of RP-NLCs,and can produce sustained-release effect.
