摘要
为了建立能够广泛应用于田间的猕猴桃细菌性溃疡病快速检测技术,参考国外经验,设计并优化LAMP反应的检测体系,并可将检测结果可视化;同时,测试评价优化后的检测体系的灵敏性及特异性;通过采集猕猴桃不同部位的组织样品,验证优化后的检测体系的田间实际应用效果。结果表明:该方法能够特异性地检测出Psa病原菌,检测灵敏度达到100 CFU/mL。经对田间样品检测测试,表明该方法田间应用效果好。
In order to establish a rapid detection technology which can be widely used for detecting bacterial canker of kiwifruit in the field,LAMP reaction with visualized results was designed and optimized based on the published methods.The sensitivity and specificity of the optimized detection system were evaluated.By collecting tissue samples from different parts of kiwifruit orchards,the field application effect of the system was verified.The results showed that the method could specifically detect the pathogenic bacteria of Pseudomonas syringae pv.actinidiae(Psa)with the detection sensitivity of 100 CFU/mL.The results of field sample testing showed that the method was also effective in the field.
