熟地黄标准汤质量标准及高效液相色谱指纹图谱研究 被引量:2

摘要 目的探讨熟地黄标准汤的质量标准,并建立其高效液相色谱(HPLC)指纹图谱。方法采用紫外-可见分光光度法测定总多糖含量;采用HPLC法测定地黄苷D含量,色谱柱为Diamonsil C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸溶液(5∶95,V/V),流速为1.0 m L/min,检测波长为203 nm,柱温为23℃,进样量为10μL。以地黄苷D为参照,测定15批熟地黄标准汤的HPLC图谱,采用"中药色谱指纹图谱相似度评价系统(2004A版)"进行相似度评价,确定共有峰。结果15批熟地黄标准汤的干膏率平均值为67.01%,总多糖含量平均值为3.58%,其HPLC图谱有2个共有峰,相似度均大于0.95。结论所建立的质量标准及HPLC指纹图谱可为熟地黄标准汤的质量评价提供参考。 Objective To investigate the quality standard of Rehmanniae Radix Praeparata standard decoction,and to establish its high-performance liquid chromatography(HPLC)fingerprint.Methods The total polysaccharide content was determined by ultraviolet-visible spectrophotometry.HPLC method was used for the content determination of rehmannioside D,the chromatographic column was Diamonsil C18 column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.1%phosphoric acid solution(5∶95,V/V),the flow rate was 1.0 m L/min,the detection wavelength was 203 nm,the column temperature was 23℃,and the injection volume was 10μL.The HPLC chromatograms of 15 batches of Rehmanniae Radix Praeparata standard decoction were determined with rehmannioside D as the reference,the Similarity Evaluation System for Chromatographic Fingerprint of traditional Chinese medicine(Version 2004 A)was used to evaluate the similarity and determine the common peaks.Results The average dry extract rate of the 15 batches of Rehmanniae Radix Praeparata standard decoction was 67.01%,and the average content of total polysaccharide was 3.58%.There were two common peaks in its HPLC chromatograms,and the similarity of fingerprints was greater than 0.95.Conclusion The established quality standard and HPLC fingerprint can provide a reference for the quality evaluation of Rehmanniae Radix Praeparata standard decoction.
