摘要
目的研究长链非编码RNA浆细胞瘤转化迁移基因1(lncRNA PVT1)在非小细胞肺癌(NSCLC)表达及与铂类化疗敏感性的关系。方法选择2012年1月-2015年6月焦作市人民医院收治的84例NSCLC患者病理活检或手术中肺癌及癌旁组织,采用实时定量PCR(RT-qPCR)检测NSCLC及癌旁组织中lncRNA PVT1表达水平,KaplanMeier法分析其与患者5年生存率的关系,Cox分析影响患者不良预后的危险因素。结果 NSCLC组织中lncRNA PVT1表达水平显著高于癌旁组织[(7.35±1.26)vs.(1.02±0.14),P<0.05]。lncRNA PVT1表达水平与吸烟史、TNM分期及淋巴结转移有关(P均<0.05)。lncRNA PVT1高表达组患者顺铂治疗有效率(28.00%vs. 55.88%)及5年总生存率(28.00%vs. 58.82%)显著低于低表达组(P均<0.05)。铂类耐药、淋巴结转移、lncRNA PVT1高表达是影响NSCLC患者不良预后的独立危险因素(P均<0.05)。结论 lncRNA PVT1在NSCLC组织中高表达,与患者铂类耐药、TNM分期、淋巴结转移及5年生存率有关,是影响NSCLC患者不良预后的危险因素。
Objective To study the expression of long non-coding RNA plasmacytoma variant translocation gene 1(lncRNA PVT1) in non-small cell lung cancer(NSCLC) and its relationship with platinum chemosensitivity. Methods From January 2012 to June 2015,84 NSCLC patients with lung cancer and paracancerous tissue were selected. Real-time quantitative PCR(RT-qPCR)was used to detect the expression of lncRNA PVT1 in NSCLC and paracancerous tissues;Kaplan Meier method was used to analyze its relationship with 5-year survival rate,and Cox analysis of the risk factors for poor prognosis. Results The expression level of lncRNA PVT1 in NSCLC was significantly higher than that in paracancerous tissues[(7.35±1.26)vs.(1.02±0.14),P<0.05]. The expression level of lncRNA PVT1 was related to smoking history,TNM stage and lymph node metastasis(P all<0.05). The effective rate of cisplatin(28.00% vs. 55.88%)and 5-year overall survival rate(28.00% vs. 58.82%) in the high expression group were significantly lower than that in the low expression group(P<0.05). Platinum resistance,lymph node metastasis and high expression of lncRNA PVT1 were independent risk factors for the poor prognosis of NSCLC patients(P all < 0.05). Conclusion LncRNA PVT1 was highly expressed in NSCLC,which was related to platinum resistance,TNM stage,lymph node metastasis and 5-year survival rate,and it was a risk factor for the poor prognosis of NSCLC patients.
