摘要
目的建立桃核承气汤的HPLC指纹图谱,并研究其与抗炎药效的谱效关系。方法采用HPLC建立15批桃核承气汤的指纹图谱,并进行药效试验,以TNF-α的抑制率和IL-10的促进率为抗炎作用的药效指标,使用灰色关联度分析法构建谱效关系。结果建立了桃核承气HPLC指纹图谱,确定了22共有峰并指认了其中9个色谱峰。药效试验结果显示,各批次桃核承气汤均具有一定的抗炎作用。经灰色关联度分析可知,除2号峰外各共有峰与2个抗炎药效指标的关联度均>0.6,其中4、10(大黄酸-8-O-β-D葡萄糖苷)、16、21(甘草酸)号峰与2个抗炎药效指标的关联度均>0.75。结论通过谱效关系研究明确了桃核承气汤抗炎药效作用是多种成分共同作用的结果。
Objective To establish the HPLC fingerprint of Taohe Chengqi decoction, and to study its relationship with anti-inflammatory effect.Methods The fingerprint of 15 batches of Taohe Chengqi decoction was established by HPLC,and the pharmacodynamic test was conducted. The inhibitory rate of TNF-α and the promotion rate of IL-10 were used as the pharmacodynamic indexes for the anti-inflammatory effect, and the spectral effect relationship was established by using the grey relational analysis method. Results HPLC fingerprint of Taohe Chengqi decoction was established, 22 common peaks were confirmed and 9 of them were identified. The results of pharmacodynamic test showed that all batches of Taohe Chengqi decoction had certain anti-inflammatory effect. According to the gray correlation analysis, the correlation between the two anti-inflammatory pharmacodynamic indexes and the common peaks except the No. 2 peak was > 0.6, and the correlation between the peaks of 4, 10(rhein-8-O-β-D glucoside), 16, 21(glycyrrhizic acid) and the two anti-inflammatory pharmacodynamic indexes was > 0.75. Conclusion Through the study of spectrum effect relationship, it is clear that the anti-inflammatory effect of Taohe Chengqi decoction is the result of the joint effect of various components.
