药桑不同部位中1-脱氧野尻霉素的提取工艺优化及含量测定分析 被引量:1

摘要 [目的]采用高效液相色谱-荧光法测定药桑不同部位1-脱氧野尻霉素(1-DNJ)含量。[方法]用AQC为衍生试剂进行衍生化反应,流动相为乙腈-乙酸铵(85∶15),流速1.0 mL/min,进样量10μL,最大激发波长250 nm,最大发射波长为395 nm下进行荧光检测;用正交试验考察最佳提取工艺条件。[结果]通过正交试验,得到最佳提取工艺为65%乙醇、料液比1∶150、超声功率150 W、提取次数2次、超声时间10 min,在最佳提取工艺下,桑叶、果实、桑枝中1-DNJ的含量分别是1.069%、0.341%、0.078%。[结论]高效液相色谱-荧光检测法操作简便、灵敏度高、准确可靠,可用于药桑不同部位的1-DNJ含量测定。 [Objective]To determine the content of 1-DNJ in different parts of mulberry by high performance liquid chromatography-fluorescence method.[Method]AQC was used as the derivative reagent to conduct the derivative reaction and generate the fluorescence product.The fluorescence detection was performed by HPLC at the maximum excitation wavelength of 250 nm and the maximum emission wavelength of 395 nm.The optimal mobile phase was ammonium acetonitrile acetate(85∶15),flow rate of 1.0 mL/min,and sample injection volume of 10μL.Orthogonal experiment was used to investigate the optimal extraction process conditions.[Result]Through orthogonal experiment,the best extraction process was 65%ethanol,solid-liquid ratio 1∶150,ultrasonic power 150 W,extraction times 2 times,ultrasonic time 10 min.Under the best extraction process,the content of 1-DNJ in mulberry leaves,fruits,and mulberry branches was 1.069.%,0.341%,0.078%,respectively.[Conclusion]The high performance liquid chromatography-fluorescence detection method was simple,sensitive,accurate and reliable,and could be used for the determination of 1-DNJ content in different parts of mulberry.
