摘要
目的探讨Wilms(WT1)基因、裸角质膜同源蛋白(NKD1)基因在急性髓系白血病中的表达及其与预后的关系。方法将2018年2月至2019年6月该院收治的100例急性髓系白血病患者纳入研究作为观察组,选取同期于该院治疗的造血系统非恶性肿瘤(缺铁性贫血)患者100例作为对照组。纳入研究者均进行WT1、NKD1基因检测,比较两组人群NKD1、WT1基因的表达水平;使用受试者操作特征曲线(ROC曲线)分析WT1、NKD1基因检测用于急性髓系白血病诊断的价值;统计患者12个月内的生存率及生存时间,采用Pearson相关分析WT1、NKD1基因表达与患者生存预后的关系。结果观察组WT1、NKD1基因表达水平均高于对照组(P<0.05)。ROC曲线分析显示,WT1基因检测的曲线下面积(AUC)为0.979,NKD1基因的AUC为0.984,两者均具有较高的诊断价值。观察组100例患者总生存率为88.00%(88/100),平均生存时间为(10.42±2.69)月,WT1、NKD1基因表达均与患者生存时间呈正相关(r=0.417、0.492,P<0.001)。结论WT1、NKD1基因表达与急性髓系白血病发生、发展密切相关。
Objective To explore the expression of Wilms(WT1)gene and naked cuticle drosophila 1(NKD1)gene in acute myeloid leukemia and their relationship with prognosis.Methods A total of 100 patients with acute myeloid leukemia admitted to the hospital from February 2018 to June 2019 were enrolled in the study as the observation group,and 100 patients with non-malignant diseases of the hematopoietic system(iron deficiency anemia)treated in this hospital during the same period were enrolled as the control group.The WT1 and NKD1 gene detection were performed to compare the expression levels of NKD1 and WT1 genes in the two groups;receiver operating characteristic curves(ROC curves)were used to analyze the value of WT1 and NKD1 gene detection in the diagnosis of acute myeloid leukemia.The survival rate and survival time of patients within 12 months were counted,and the relationship between WT1 and NKD1 gene expression and survival prognosis was analyzed by Pearson correlation.Results The expression levels of WT1 and NKD1 genes in the observation group were higher than those in the control group(P<0.05).ROC curve analysis showed that the area under the curve(AUC)of WT1 gene detection was 0.979,and the AUC of NKD1 gene was 0.984,both of which have high diagnostic value.The overall survival rate of 100 patients was 88.00%(88/100),and the average survival time was(10.42±2.69)months.The expression of WT1 and NKD1 genes were positively correlated with the survival time of patients(r=0.417,0.492,P<0.001).Conclusion The expression of WT1 and NKD1 genes is closely related to the occurrence and development of acute myeloid leukemia.
