摘要
目的:探究微小RNA-21(miR-21)、微小RNA-29a(miR-29a)在冠心病(CHD)患者血清中的水平及与心电图QRS波时限的相关性。方法:收集162例CHD患者作为CHD组,并选取同期166例健康体检者作为对照组。采用实时荧光定量PCR(qRT-PCR)法检测两组血清miR-21、miR-29a水平。以酶联免疫吸附法(ELISA)测定基质金属蛋白酶-2(MMP-2)、肿瘤坏死因子-α(TNF-α)水平。使用心电图机检测QRS波时限。分析CHD患者血清miR-21、miR-29a、QRS波时限与MMP-2、TNF-α,以及miR-21、miR-29a与QRS波时限的相关性。采用Logistic回归模型分析CHD影响因素。结果:CHD组三酰甘油、总胆固醇、低密度脂蛋白、miR-21、miR-29a、QRS波时限、MMP-2、TNF-α水平高于对照组,高密度脂蛋白低于对照组(均P<0.05)。CHD患者血清miR-21、miR-29a、QRS波时限与MMP-2、TNF-α,以及miR-21、miR-29a表达水平与QRS波时限呈正相关(均P<0.05)。miR-21、miR-29a、QRS波时限、MMP-2、TNF-α均是影响CHD发生的危险因素(均P<0.05)。结论:CHD患者血清miR-21、miR-29a呈高表达,两者均与心电图QRS波时限呈正相关。miR-21、miR-29a可能与心功能不全相互影响,从而在CHD发病过程中发挥重要作用。
Objective:To investigate the levels of serum microRNA-21(miR-21)and microRNA-29a(miR-29a)in patients with coronary heart disease(CHD)and their correlations with QRS duration of ECG.Methods:162 CHD patients were collected as the CHD group,and 166 healthy physical examination patients during the same period were selected as the control group.The levels of serum miR-21 and miR-29a were detected by real-time fluorescence quantitative PCR(qRT-PCR).The levels of matrix metalloproteinase-2(MMP-2)and tumor necrosis factor-α(TNF-α)were measured by enzyme-linked immunosorbent assay(ELISA).The QRS duration was measured by electrocardiograph in CHD patients.The correlations between serum miR-21,miR-29a,QRS duration and MMP-2,TNF-α,between miR-21,miR-29a and QRS duration were analyzed.The influencing factors of CHD were analyzed by Logistic regression.Results:The levels of triglyceride,total cholesterol,low density lipoprotein,miR-21,miR-29a,QRS duration,MMP-2 and TNF-αin the CHD group were higher than those in the control group(all P<0.05),and high density lipoprotein was lower than that in the control group(P<0.05).In CHD patients,the levels of serum miR-21,miR-29a and QRS duration were positively correlated with the levels of MMP-2 and TNF-α,and the levels of miR-21 and miR-29a were positively correlated with QRS duration(all P<0.05).The levels of miR-21,miR-29a,QRS duration,MMP-2 and TNF-αwere all risk factors for CHD(all P<0.05).Conclusion:The levels of serum miR-21 and miR-29a in CHD patients are high,and both of them are positively correlated with QRS duration.The miR-21 and miR-29a may interact with cardiac insufficiency and play an important role in the pathogenesis of CHD.
