摘要
扫描电镜(SEM)和透射电镜(TEM)是用于微观分析的重要研究工具。但在教学过程中,发现不少本科生缺乏电镜类仪器的基础知识,常将SEM和TEM混为一谈,实验目的性不明确,致使仪器的实际使用效率偏低。结合笔者多年电镜管理经验,从工作原理、制样要求、样品测量、图像拍摄、注意事项等方面分析了TEM和SEM的差别,结合若干具体案例,描述了两种电镜用于微观分析的主要测试功能及各自特点,并就如何提高TEM和SEM的使用效率,提出针对本科生举办电镜类大型仪器讲座、实施预实习培训制度、加强仪器使用的监督和管理等建议。
The scanning electron microscope(SEM)and transmission electron microscope(TEM)are the most important instruments used to microstructure analysis.During the teaching process,quite a few students have been found the deficiency in the foundation on electro-microscopy.They often confuse SEM with TEM,and mindlessly follow with other students in the experiments,resulting in the low service efficiency of electro-microscopy.Combined with the many years of experience gained in the electro-microscopy management,the difference between the TEM and SEM was explored from such aspects as principle of work,requirements for sample preparation,measurement of sample,snapping of sample image,and points for attention.Many samples were presented and the characteristics of two kinds of electro-microscopy and their measuring functions were described.Three suggestions on how to raise the service efficiency of TEM and SEM were proposed,including arrange electro-microscopy lectures for undergraduates,implement previewing and training regulation,and strengthen supervision and management on the use of instruments.
