硫酸镁对高氧脑损伤新生大鼠的神经保护作用 被引量:2

摘要 目的探讨硫酸镁对高氧性脑损伤新生大鼠的神经保护作用。方法将30只6日龄SD大鼠随机分为对照组、高氧组、硫酸镁组,每组10只。将高氧组、硫酸镁组新生大鼠暴露于氧气浓度约为80%的氧箱中24 h,建立高氧脑损伤模型。建模成功后,硫酸镁组大鼠在生后第7、8、9天分别给予50 mg/kg硫酸镁腹腔内注射。于日龄10 d、14 d各处死每组5只新生大鼠,取大鼠脑组织检测谷氨酸、γ-氨基丁酸(GABA)、过氧化脂质(LOP)浓度以及白细胞介素1(IL-1)mRNA相对表达水平。结果高氧组谷氨酸和LPO浓度、谷氨酸/GABA比值、IL-1 mRNA相对表达水平均高于对照组,硫酸镁组以上指标均低于高氧组(均P<0.05)。结论硫酸镁可抑制脑组织兴奋性氨基酸的产生、氧化应激反应和炎症反应,对高氧性脑损伤新生大鼠有神经保护作用。 Objective To study the neuroprotective effect of magnesium sulfate on hyperoxia-induced brain damage in neonatal rats.Methods Thirty SD rats aged 6 days were randomly divided into control group,hyperoxia group and magnesium sulfate group,with 10 rats in each group.Neonatal rats in the hyperoxia and magnesium sulfate groups were exposed to 80%concentration of oxygen in the tank for 24 hours to establish a model of hyperoxia-induced brain damage.After successful modeling,rats in the magnesium sulfate group were intraperitoneally injected with magnesium sulfate of 50 mg/kg on days 7,8,and 9 after birth.Five neonatal rats in each group were sacrificed on days 10 and 14 after birth separately,and the brain tissue of the rats were removed to measure the concentrations of glutamic acid(GLU),γ-aminobutyric acid(GABA)and lipid peroxide(LOP),as well as relative expression level of interleukin 1(IL-1)mRNA.Results The concentrations of GLU and LPO,GLU to GABA ratio,and relative expression level of IL-1 mRNA in the hyperoxia group were higher than those in the control group,whereas the magnesium sulfate group exhibited lower aforementioned indicators than the hyperoxia group(all P<0.05).Conclusion Magnesium sulfate can inhibit production of brain tissue excitatory amino acid,oxidative stress response and inflammatory response,and has a neuroprotective effect on neonatal rats with hyperoxia-induced brain damage.
