急性冷应激对断奶仔猪全血中HSP70/TLR4/NF-κB通路的影响 被引量:3

Effect of acute cold stress on HSP70/TLR4/NF-κB pathway in whole blood of weaned piglets

摘要 为了探讨急性冷应激对断奶仔猪全血中HSP70/TLR4/NF-κB通路关键分子表达的影响,将16头30日龄的断奶仔猪随机分成常温对照组,冷应激2,6,12h组,每组4头。常温对照组饲养温度为(24±2)℃,冷应激组饲养温度为(4±2)℃。采集4组仔猪的全血,qRT-PCR用于检测分析4组仔猪全血中热休克蛋白70(HSP70)、Toll样受体4(TLR4)、髓样分化因子88(MyD88)、核转录因子-κB(NF-κB)RelA(p65)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)和白介素-10(IL-10)的表达变化。结果显示,与常温对照组相比,冷应激2h组断奶仔猪全血中HSP70水平极显著升高,TLR4、IL-6水平显著升高,MyD88、p65水平极显著降低,TNF-α、IL-10水平与常温对照组相比无明显差异;冷应激6h组HSP70、TLR4、TNF-α、IL-6水平极显著升高,MyD88、p65、IL-10水平极显著降低;冷应激12h组MyD88、p65水平极显著升高,IL-10水平显著降低,HSP70、TLR4、TNF-α、IL-6水平与常温对照组相比无明显差异。结果表明,急性冷应激会导致断奶仔猪全血中HSP70/TLR4/NF-κB通路被激活,引起先天性免疫应答和炎症反应,随着冷应激时间的增加,免疫功能下降。可以考虑把HSP70/TLR4/NF-κB通路关键分子作为评价断奶仔猪遭受冷应激程度的客观指标。 To explore the effect of acute cold stress on the expression of key molecules of HSP70/TLR4/NF-κB pathway in whole blood of weaned piglets,1630-day-old weaned piglets were randomly divided into the normal temperature control group,2hcold stress group,6hcold stress group and 12hcold stress group,4heads in each group.The rearing temperature of the normal temperature control group was(24±2)℃,and the rearing temperature of the cold stress group was(4±2)℃.Then the whole blood of 4groups of piglets was collected,and qRT-PCR was used to detect and analyze the changes of the expression of HSP70,TLR4,MyD88,p65,TNF-α,IL-6and IL-10in the whole blood.The results showed that compared with the normal temperature control group,the levels of HSP70in the whole blood of the weaned piglets were significantly increased in the 2hcold stress group,the levels of TLR4and IL-6were significantly increased,the levels of MyD88and p65were significantly decreased,and the levels of TNF-αand IL-10were not significantly different from those in the normal temperature control group.In the 6hcold stress group,the levels of HSP70,TLR4,TNF-αand IL-6 were significantly increased,and the levels of MyD88,p65,and IL-10were significantly decreased.In the 12hcold stress group,the levels of MyD88and p65were significantly increased,the levels of IL-10were significantly decreased,and the levels of HSP70,TLR4,TNF-α,and IL-6were not significantly different from those in the normal temperature control group.From the above results,it is concluded that acute cold stress can cause the HSP70/TLR4/NF-κB pathway to be activated in whole blood of weaned piglets,causing innate immune responses and inflammatory responses,and immune function decreases with the extension of cold stress time.Therefore,the key molecules of HSP70/TLR4/NF-κB pathway can be considered as objective indicators to evaluate the degree of cold stress on weaned piglets.
