摘要
以花生红衣为研究对象,采用单因素试验及正交设计优化花生红衣中原花青素的提取工艺,通过大孔树脂吸附对原花青素进行分离纯化,并对提纯产物的体外抗氧化活性进行了初步研究。结果表明,以香草醛-盐酸法测定原花青素含量为评价指标,根据单因素试验结果设计正交试验,得到原花青素最佳提取工艺参数为乙醇体积分数70%,超声温度30℃,超声时间17.5 min,提取次数3次,料液比1∶16 (g:mL),得率为7.82%±0.02%。在小型离子交换柱中,采用AB-8型大孔吸附树脂为柱填料,以乙醇为洗脱剂,进行分离纯化,经冻干后得到干物质中原花青素的纯度为76.85%±0.24%,较未纯化前提升42.57%。原花青素对DPPH自由基清除能力最好,当质量浓度为0.05 mg/m L时清除率达到最大值为92.08%±0.01%。以上结果表明,花生红衣中的原花青素具有一定的抗氧化活性,是一种潜在的天然抗氧化剂。
Taking peanut red coat as the research object,single-factor and orthogonal design was used to optimize the extraction process of proanthocyanidins in peanut red coat.The procyanidins were separated and purified by macroporous resin adsorption,and the in vitro antioxidant activity of the purified product was preliminary the study.The results showed that the vanillin-hydrochloric acid method was used to determine the content of proanthocyanidins,and the orthogonal experiment was designed according to the single-factor experiment results.The optimal extraction process parameters of proanthocyanidins were obtained:ethanol concentration was 70%,ultrasonic temperature was 30℃,ultrasonic time was 17.5 min.The extraction times were 3 times,the ratio of material to liquid was 1∶16(g∶mL),and the yield was 7.82%±0.02%.In a small ion exchange column,the macroporous adsorption resin AB-8 was used as the column packing,and ethanol is used as the eluent for separation and purification.After freeze-drying,the purity of the proanthocyanidin in the dry matter was 76.85%±0.24%,which is relatively unpurified.The previous increase was 42.57%.Proanthocyanidins have the best scavenging ability for DPPH free radicals.When the concentration was 0.05 mg/mL,the scavenging rate reaches the maximum of 92.08%±0.01%.The above results indicate that the proanthocyanidin in the peanut red coat had certain antioxidant activity and was a potential natural antioxidant.
