摘要
目的:探讨紫云英苷(astragalin)对弥漫性大B细胞淋巴瘤(DLBCL)细胞系OCI-LY8的作用及机制。方法:将0~125μg/L的astragalin作用于OCI-LY8细胞,采用CCK-8法检测细胞活力,Hoechst 33342染色和流式细胞术分别分析OCI-LY8细胞核型变化和凋亡率,利用RT-qPCR和Western blot检测OCI-LY8细胞凋亡相关蛋白和JAK/STAT信号通路相关蛋白的表达水平,小鼠抑瘤实验观察紫云英苷对OCI-LY8细胞生长的作用。结果:紫云英苷可显著抑制OCI-LY8细胞活力,诱导其凋亡,促进p53、caspase-3和Bax的mRNA和蛋白病毒,抑制Bcl-2的mRNA和蛋白表达,降低JAK1、JAK2和STAT3的磷酸化水平。结论:紫云英苷可通过上调p53表达和影响JAK/STAT信号通路来诱导OCI-LY8细胞凋亡,在DLBCL治疗中可能发挥一定的作用。
AIM:To explore the effect of astragalin on diffuse large B-cell lymphoma(DLBCL)cell line OCILY8.METHODS:The OCI-LY8 cells were treated with 0-125μg/L astragalin.CCK-8 assay was used to measure the cell viability,and Hoechst 33342 staining and flow cytometry were used to analyze the karyotype changes and apoptosis.RT-qPCR and Western blot were used to determine the expression levels of apoptosis-and JAK/STAT sinaling pathway-re-lated proteins.The anti-tumor experiment in mice was used to confirm the anti-cancer effect of astragalin on the growth of OCI-LY8 cells.RESULTS:Astragalin significantly inhibited the viability of OCI-LY8 cells,and induced their apopto-sis.Astragalin promoted the expression of p53,caspase-3 and Bax at mRNA and protein levels,inhibited the expression of Bcl-2 at mRNA and protein levels,and attenuated the phosphorylation of JAK1,JAK2 and STAT3.CONCLUSION:Astragalin induces apoptosis of OCI-LY8 cells by up-regulating p53 expression and influencing JAK/STAT signaling path-way,which may play a certain part in the treatment of DLBCL.
