摘要
目的研究分析丹参DNA提取方法的优化。方法针对丹参DNA提取方法展开相应研究,选取盆栽陕西商洛一年生紫花丹参的叶片为研究材料,比较4种方法提取丹参DNA的效果。结果改良版CTAB法的DNA提取质量大于SDS法2、CTAB法、SDS法1(P<0.05)。四种DNA提取方法提取的丹参DNA浓度、纯度存在明显差异(P<0.05)。结论改良版CTAB法提取丹参DNA的效果最好,操作步骤简单,值得推广应用。
Objective To study and analyze the optimization of DNA extraction methods of Salvia miltiorrhiza.Methods The corresponding research was carried out on the DNA extraction methods of Salvia miltiorrhiza,and the leaves of potted annual purple Salvia miltiorrhiza in Shangluo,Shaanxi were selected as the research materials,and the effects of 4 methods for extracting the DNA of Salvia miltiorrhiza were compared.Results The DNA extraction quality of the modified CTAB method was better than that of SDS method 2,CTAB method,and SDS method 1(P<0.05).There were significant differences in the concentration and purity of Salvia miltiorrhiza DNA extracted by the four DNA extraction methods(P<0.05).Conclusion The improved CTAB method has the best effect on DNA extraction of Salvia miltiorrhiza with simple steps and is worthy of popularization and application.
