松脂醇二葡萄糖苷促进体外培养成骨细胞骨形成的作用研究 被引量:7

摘要 目的探讨松脂醇二葡萄糖苷对体外培养成骨细胞骨形成的作用。方法应用分次酶消化法得到大鼠乳鼠颅骨成骨细胞,随机分为空白组和实验组,通过不同浓度松脂醇二葡萄糖苷处理检测成骨细胞碱性磷酸酶活性,确定最佳作用浓度后行钙化结节染色,检测成骨细胞成骨相关蛋白和基因表达情况。结果松脂醇二葡萄糖苷处理成骨细胞的最佳作用浓度为1×10^(-6) mol/L(P<0.01)。1×10^(-6) mol/L松脂醇二葡萄糖苷处理成骨细胞后钙化结节形成程度明显高于空白组。与空白组相比,1×10^(-6) mol/L松脂醇二葡萄糖苷处理后成骨细胞BMP-2、RUNX-2和OSX基因和蛋白表达显著升高(P<0.05或P<0.01)。结论1×10^(-6) mol/L松脂醇二葡萄糖苷可以促进体外培养成骨细胞的骨形成,增加钙化结节面积,并可升高成骨相关蛋白基因表达水平。 Objective To investigate effect of Pinoresinol Diglucoside on bone formation of osteoblasts in vitro culture.Methods The calvarial osteoblasts of neonate rats were obtained by split-enzyme digestion and randomly divided into blank group and experimental group.Alkaline phosphatase(ALP)activities of osteoblasts were detected after treatment by different concentrations of Pinoresinol Diglucoside.After the optimal concentration was determined,the calcified nodule staining was performed to detect expressions of osteogenic-related genes and proteins in osteoblasts.Results The optimal concentration of Pinoresinol Diglucoside was 1×10^(6) mol/L(P<0.01).After osteoblasts were treated with 1×10^(6) mol/L Pinoresinol Diglucoside,the degree of calcified nodule formation was significantly higher than that in blank group.Compared with those in blank group,gene and protein expressions of BMP-2,RUNX-2 and OSX in osteoblasts after treatment by 1×10^(6) mol/L Pinoresinol Diglucoside were significantly increased(P<0.05 or P<0.01).Conclusion The 1×10^(6) mol/L Pinoresinol Diglucoside may promote bone formation of osteoblasts in vitro culture,enlarge the area of calcified nodules and increase expressions of osteogenic-related genes and proteins.
