芭蕉根促成骨细胞增殖及分化的有效组分筛选研究 被引量:6

摘要 目的对芭蕉根提取物各组分促成骨细胞增殖及分化的有效组分进行筛选。方法于2018年6月—2019年1月,采用人成骨肉瘤SaOS-2细胞,芭蕉根各组分组加入浓度为100 mg/L作用细胞48 h后,用噻唑蓝(MTT)法检测细胞增殖,酶联免疫法(ELISA)法检测细胞上清液中碱性磷酸酶(ALP)、钙离子(Ca^2+)和骨钙素(BGP)的含量。结果与空白对照组比较,药物B、C、D组分均可显著促进细胞的增殖(P<0.05),分别使细胞增殖16.74%、5.57%、10.02%;升高细胞中ALP的含量(P<0.005),分别使细胞中ALP的含量增加58.11%、21.45%、39.57%;促进细胞中Ca^2+、BGP的分泌(P<0.01),其中B组分的作用最显著。结论B组分为芭蕉根促成骨细胞增殖及分化的有效组分,其可能通过升高ALP、Ca2+的含量、促进BGP的分泌从而促成骨细胞增殖及分化。 Objective To screen the effective components of the growth and differentiation of bone cells in the extracts of banana root. Methods From June 2018 to January 2019, human osteosarcoma SaOS-2 cells were used. The cells of the banana roots were added to the cells treated with 100 mg/L for 48 h, and then detected by MTT assay. Cell proliferation and enzyme-linked immunosorbent assay (ELISA) were used to detect the contents of alkaline phosphatase (ALP), calcium (Ca^2+) and osteocalcin (BGP) in the supernatant. Results Compared with the blank control group, the B, C, and D components significantly promoted cell proliferation (P<0.05), and the cells proliferated 16.74%, 5.57%, and 10.02%, respectively. The content of ALP (P<0.005) increased the content of ALP in cells by 58.11%, 21.45% and 39.57%, respectively, and promoted the secretion of Ca^2+ and BGP in cells (P<0.01). Most notable in group B. Conclusion Group B of component is an effective component of banana root promoting osteoblast proliferation and differentiation. It may promote osteoblast proliferation and differentiation by increasing the content of ALP and Ca^2+ and promoting the secretion of BGP.
