摘要
目的 探讨高效液相色谱法在检测中成药物中糖精钠含量的应用效果.方法 应用安捷伦1260高效液相色谱系统,配备可变波长紫外检测器,其色谱条件为:C18色谱柱(150mm×4.6mm,5μm),检测波长为230nm;流动相为甲醇-乙酸铵溶液(7.5:92.5);流速为0.8mL/min;柱温35℃.检测含不同浓度糖精钠的5份样品.结果 应用高效液相色谱法对5份样品进行测定,糖精钠含量在0.05mg/g~0.80mg/g的范围内,测量结果有较高的准确度.在同一份样品中分别添加10.0μg、5.0μg及2.5μg浓度的标准溶液计算出回收率,平均回收率为100.2%.结论 高效液相色谱法在检测中成药物中的糖精钠含量具有较高的准确度和回收率,而且该方式操作比较方便,在临床药物检验中值得大力推广.
Objective To explore the application of high performance liquid chromatography in detecting the content of saccharin sodium in Chinese m edicines.Methods Using Agilent 1260 high performance liquid chromatography system,equipped with variable wavelength ultraviolet detector,the chromatographic conditions are:C18 column(150mm×4.6mm,5μm),detection wavelength is 230 nm;mobile phase is methanolammonium acetate solution(7.5:92.5);The flow rate is 0.8mL/min;the column temperature is 35℃.Test 5 samples of sodium saccharin at different concentrations.Results Using high performance liquid chromatography to determine the 5 samples,the content of saccharin sodium was in the range of 0.05mg/g to 0.80mg/g,and the measurement results had high accuracy.The recovery rate was calculated by adding 10.0μg,5.0μg and 2.5μg standard solutions to the same sample,and the average recovery rate was 100.2%.Conclusion High performance liquid chromatography has high accuracy and recovery rate in detecting the content of sodium saccharin in finished medicines,and this method is more convenient to operate,and it is worthy of promotion in clinical drug testing.
