Influence of Operation Conditions on Determination of Molecular Weight Distribution for Organic Matters by Combined Technology of HPSEC-UV-TOC

摘要 采用高效凝胶色谱法(HPSEC)与紫外检测器(UV)、总有机碳(TOC)在线检测仪联用测定葡聚糖(分子量为1000,T1)及天然地表水有机物相对分子质量分布。以磷酸盐缓冲盐作为流动相,考察pH、离子强度和配样溶液对测定结果的影响。结果表明:样品pH对葡聚糖T1分子量分布测定结果影响较小;离子强度对有机物相对分子质量分布的测定具有一定影响,在色谱柱允许的分子量范围内,离子强度越大,UV响应越高,而TOC峰面积越小;采用超纯水配制的样品与采用流动相配制的样品测定结果差别较小;该联用系统能够很好地表征黄河水库原水及其4种亲疏水性组分的相对分子量分布。 A high performance size exclusion chromatography(HPSEC)with UV and TOC detectors was used to analyze relative molecular weight(MW)distribution for dextran(MW 1000,T1)and natural organic matters(NOM)in surface water.Effects of ionic strength,pH value and sample solvents on the performance of the HPSEC-UV-TOC using phosphate buffer as mobile phase were also investigated.It was found that the sample pH value had minor influence on determination of molecular weight distribution for dextran T1.Effect of ionic strength on determination of relative molecular weight distribution for organic matters was relelatively notable.It was shown that the higher ionic strength caused increased UV peak response and decreased TOC peak area within the available MW range of chromatographic column.Determination results of the samples prepared by ultrapure water were similar to that of the sample solvents adopted as mobile phase.HPSEC-UV-TOC system could be used accurately to characterize relative molecular weight distribution and its four fractions in the Yellow River.
