摘要
通过高效液相色谱-蒸发光散射检测仪建立三氯蔗糖及其相关物质的含量的检测方法。配置不同浓度的三氯蔗糖溶液(0.0005%、0.001%、0.002%、0.005%、0.01%、0.02%、0.05%、0.1%、0.2%),采用C18色谱柱,蒸发光散射检测器,甲醇-水为流动相进行梯度洗脱,柱温35℃,雾化器温度75℃,空气流量2.5L/min,得到不同浓度三氯蔗糖溶液的峰面积,取浓度与峰面积的双对数发现三氯蔗糖溶液在0.0005%~1%之间内呈现良好的线性关系,得到回归方程y=0.7364x-5.0742(R^(2)=0.9996),将样品通过高效液相色谱-蒸发光散射检测法得到三氯蔗糖及一些相关物质的峰面积,代入此方程得到浓度,与稀释比例乘积得到三氯蔗糖及其相关物质的含量。
A HPLC-ELSD method was established to determine sucralose and its related substances.Different concentrations of sucralose solution(0.0005%,0.001%,0.002%,0.005%,0.01%,0.02%,0.05%,0.1%,0.2%)were made.Detecting parameters were:C18 chromatographic columns and ELSD,Methanol-water as a mobile phase gradient elution,Column temperature 35℃,atomizer 75℃,air flow of 2.5 L/min.Peak areas of sucralose solution with different concentration were caculated.Using double logarithm of concentration and peak area,it was found that sucrose solution showed a good linear relationship between 0.0005%and 1%.The regression equation was y=0.7364x-5.0742(R^(2)=0.9996).The concentration can be obtained by substitute of peak area of sucralose and some related substances obtained by HPLC-ELSD into the equation,the final content of sucralose and its related substances then can be calculated by multiple of dilution ratio.
