哈蟆油与牛蛙油蛋白质组学差异分析 被引量:1

摘要 目的旨在筛选哈蟆油和牛蛙油中特异性的蛋白质,发现这些差异蛋白与生长环境、冬眠及发育的关系。方法采用iTRAQ结合质谱技术建立哈蟆油和牛蛙油蛋白质组数据库,通过比较两个蛋白质组数据库,对筛选的差异蛋白进行Gene ontology功能注释和生物信息学分析。结果通过对牛蛙油与哈蟆油中蛋白质定量的数据进行差异比较,筛选到差异表达蛋白质总数为77个,其中上调蛋白质69个,下调蛋白质8个。哈蟆油中含量较高的8个差异蛋白主要参与免疫调节过程;牛蛙油中含量较高的差异蛋白主要参与生长发育、能量代谢、应激反应、骨架形成等过程。结论哈蟆油中含量较高的8个差异蛋白质可作为哈蟆油区别于牛蛙油的重要成分,同时哈蟆油的特异性膨胀度可能与含量较高的凝胶形成型黏蛋白类密切相关。 Objective To screen those differentially expressed proteins from the proteomics database of rana oviduct and bullfrog oil,and to work out further the relationship between these differentially expressed proteins and their different habitats,hibernation manners and development ways.Methods The proteomics databases of rana oviduct and bullfrog oil were established by using iTRAQ technology in combination with mass spectrometry(MS).By comparing the two proteomics databases,gene ontology function annotation and bioinformatics analysis were performed on the screened differential proteins.Results By comparing the quantitative data of proteins in bullfrog oil and rana oviduct,a total of 77 differentially expressed proteins were screened,of which 69 were up-regulated proteins and 8 down-regulated proteins.It was discovered that the eight differential proteins with a higher content in rana oviduct were mainly involved in the immune regulation process,while the differential proteins with a higher content in bullfrog oil were mainly involved in processes as growth and development,energy metabolism,stress response,and skeleton formation.Conclusion The eight differential proteins with a higher content in rana oviduct can be used as important components to distinguish rana oviduct from bullfrog oil.At the same time,the specific swelling degree of rana oviduct may be closely related to the higher content of gel-forming mucins.
