基于网络药理学探讨淫羊藿治疗良性前列腺增生症的作用机制 被引量:2

摘要 目的:探讨淫羊藿治疗良性前列腺增生症(BPH)的作用机制。方法:利用中药系统药理学分析平台数据库(TCMSP)筛选淫羊藿的有效化学成分,并收集对应的作用靶点;使用DisGeNET数据库获取BPH相关靶点,并筛选淫羊藿有效化学成分与BPH的交集靶点,采用Cytoscape 3.6.0软件和String数据库绘制“淫羊藿有效化学成分-BPH-靶点”和PPI网络图;通过DAVID数据库对核心靶点进行GO富集和KEGG通路分析。结果:筛选出淫羊藿有效化学成分22个,与BPH交集44个靶点。“淫羊藿化合物-BPH-靶点”网络中degree值前3名的核心化学成分为quercetin(槲皮素)、kaempferol(山柰酚)、luteolin(木犀草素),PPI网络中degree值前3名的核心靶点为TP53、ESR1、IL6。GO富集与KEGG通路分析主要涉及雌激素反应、腺体发育、蛋白激酶结合等过程和HIF-1、p53、PI3K-Akt、JAK-STAT、VEGF、ErbB等信号通路。结论:淫羊藿中的槲皮素、山柰酚和木犀草素等化学成分,TP53、ESR1和IL6等核心靶点,雌激素反应、腺体发育、蛋白激酶结合等过程和HIF-1、p53、PI3K-Akt、JAK-STAT、VEGF、ErbB等信号通路是淫羊藿治疗BPH的潜在作用机制,为后续深入研究其具体机制提供了新的理论基础。 In this study we investigated the mechanism of Epimedii Folium in the treatment of benign prostatic hyperplasia(BPH).TCMSP database was used to screen the effective chemical components of Epimedii Folium,and the corresponding targets were collected.The BPH related targets were obtained by using DisGeNET database.The intersectional targets of the effective chemical components of Epimedii Folium and BPH were screened out,and Cytoscape 3.6.0 and String database were used to draw the“Epimedii Folium effective chemical components-BPH-targets”and the PPI network diagrams.GO enrichment and KEGG pathway analysis of the core targets were conducted by using DAVID database.A total of 22 effective chemical components were screened from Epimedii Folium,and 44 targets were intersected with BPH.Quercetin,kaempferol and luteolin were the top three core compounds in the“Epimedii Folium compounds-BPH-targets”network,and TP53,ESR1 and IL6 were the top three core targets in the PPI network.GO enrichment and KEGG pathway analysis mainly involved processes of estrogen response,gland development,and protein kinase binding,and signaling pathways of HIF-1,p53,PI3K-Akt,JAK-STAT,VEGF,and ErbB.In conclusion,the potential mechanism of Epimedii Folium in the treatment of BPH might involve compounds of quercetin,kaempferol and luteolin,core targets of TP53,ESR1 and IL6,processes of estrogen response,gland development and protein kinase binding,and signaling pathways of HIF-1,p53,PI3K-Akt,JAK-STAT,VEGF and ErbB.This study has provided new theoretical basis for further study on the specific mechanism of Epimedii Folium in the treatment of BPH.
