摘要
目的:探讨银杏叶滴剂对口腔溃疡大鼠的作用及对抗氧化通路核因子E2相关因子(nuclear factor erythroid-2 related factor,Nrf2)/抗氧化反应元件(antioxidant response element,ARE)通路的调控作用。方法:取50只雄性SD大鼠,其中40只采用化学灼烧法建立口腔溃疡模型,随机分为模型组、银杏叶滴剂低剂量(25 mg·kg^(-1))组、银杏叶滴剂中剂量(50 mg·kg^(-1))组、银杏叶滴剂高剂量(100 mg·kg^(-1))组,其余10只为对照组,连续给药10 d。比较给药第0、1、3、7天时各组大鼠溃疡面积;给药10 d后处死大鼠,检测溃疡组织中超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、丙二醛(malondialdehyde,MDA)水平,TH、Nrf2、HO-1蛋白表达及TH mRNA、Nrf2 mRNA、HO-1 mRNA水平。结果:与模型组比较,干预第3、7天银杏叶滴剂低、中、高剂量溃疡面积均明显减小(P<0.05);银杏叶滴剂低、中、高剂量组溃疡组织中SOD、GSH-Px水平明显升高、MDA水平明显降低(P<0.05),TH、Nrf2、HO-1蛋白表达明显升高(P<0.05),TH mRNA、Nrf2 mRNA、HO-1 mRNA水平明显升高(P<0.05)。结论:银杏叶滴剂可有效促进大鼠口腔溃疡愈合,可能与激活溃疡组织内Nrf2/ARE通路改善氧化应激有关。
Objective:To explore the therapeutic effects of ginkgo biloba drops on oral ulcer rats and the regulation of nuclear factor erythroid-2 related factor 2(Nrf2)/antioxidant response element(ARE)pathway in the antioxidant pathway.Methods:Fifty male SD rats were taken,40 of which were established by chemical cauterization to establish oral ulcer models.They were randomly divided into model group,ginkgo bioba drops low-dose(25 mg·kg^(-1))group,and ginkgo bioba drops medium-dose(50 mg·kg^(-1))group,ginkgo bioba drops high-dose(100 mg·kg^(-1))group,and the remaining 10 rats were the control group.They were given continuous administration for 10 days.On the 0,1,3,and 7 days of the administration,the ulcer area of the rats in each group was compared;after 10 days of administration,the rats were sacrificed and the levels of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in the ulcer tissue,TH,Nrf2,HO-1 protein expression and TH mRNA,Nrf2 mRNA,HO-1 mRNA levels were detected.Results:Compared with the model group,the area of ulcers in the low,medium and high dose groups of ginkgo biloba drops were significantly reduced on the 3 rd and 7 th day(P<0.05);SOD and GSH-Px levels in the ulcer tissues of the low,medium and high dose groups of ginkgo biloba drops were significantly increased,MDA level was significantly decreased(P<0.05);TH,Nrf2,HO-1 protein expression was significantly increased(P<0.05),TH mRNA,Nrf2mRNA,HO-1 mRNA levels were significantly increased(P<0.05).Conclusion:Ginkgo bioba drops can effectively promote the healing of oral ulcers in rats,which may be related to activating the Nrf2/ARE pathway in the ulcer tissue to improve oxidative stress.
