摘要
目的制备一种氨基甲酸铵和磷酸盐缓冲液(PBS)复合活化电极,用于快速检测尿液中的多巴胺(DA)和尿酸(UA)。方法将裸玻碳电极(GCE)先后置于氨基甲酸铵溶液和磷酸盐缓冲溶液中,采用恒电位法进行活化,再采用循环伏安法在PBS中稳定扫描得到复合活化电极(CA-GCE)。接着使用循环伏安法、交流阻抗法对CA-GCE的电极性能进行表征。建立示差脉冲伏安法实现对DA和UA的快速灵敏检测。结果在最佳条件下,CA-GCE对DA和UA表现出优异的电催化活性。在DA和UA的浓度分别为3μmol/L~120μmol/L和3μmol/L~70μmol/L,峰电流与其浓度呈良好的线性关系,检测限(3σ/S)分别为0.15μmol/L和0.43μmol/L。CA-GCE具有良好的稳定性、选择性和重现性。结论该方法灵敏度高、检测范围广、检测速度快,适用于尿液样品中DA和UA的检测。
Objective An electrochemical sensor based on ammoniated-PBS activated electrode(CA-GCE)was prepared and used for the determination of dopamine(DA)and uric acid(UA).Methods Bare glass carbon electrode(GCE)was placed in ammonium carbamate solution and then activated by the potentiostatic method.Then,the above aminated GCE was activated in phosphate buffer solution(PBS)by simple anodic and cathodic polarization.Subsequently,cyclic voltammetry and electrochemical impedance spectroscopy were used to characterize the electrode performance of CA-GCE.Differential pulse voltammetry was used to determine the concentration of DA and UA.Results Under the optimal conditions,CA-GCE showed excellent electrocatalytic activity on DA and UA.The linear detection ranges for DA and UA were within 3μmol/L-120μmol/L and within 3μmol/L-70μmol/L,with their detection limits(3σ/S)of 0.15μmol/L and 0.43μmol/L,respectively.CA-GCE has good stability,selectivity and reproducibility.Conclusion The method has high sensitivity,wide detection range and fast detection speed,and is used for the detection of DA and UA in urine samples.
