摘要
目的:探究传统中药葛根素对腰椎间盘突出症引起的神经根性疼痛的作用及机制。方法:用自体髓核(NP)移植的方法复制大鼠腰椎间盘突出症模型,连续7天腹腔注射葛根素(100 mg·kg^(−1)·d^(−1))观察镇痛效果;采用机械性撤足阈值(PWT)和热撤足潜伏期(PWL)作为痛觉阈值检测指标;Western blot方法检测脊髓组织TLR4和p-NF-κB(p-p65)的蛋白水平;免疫荧光染色法观察脊髓小胶质细胞的激活情况;ELISA法检测脊髓组织中相关致炎和抗炎细胞因子的含量。结果:葛根素显著缓解NP移植引起的痛觉过敏和痛觉超敏;葛根素降低了脊髓组织中TLR4和p-p65的蛋白水平,抑制了脊髓小胶质细胞的激活;此外,葛根素还降低了脊髓组织中致炎细胞因子IL-1β、IL-6和TNF-α的含量,增加了抗炎细胞因子IL-10的含量。结论:葛根素可通过抑制小胶质细胞TLR4/NF-κB介导的炎症反应治疗腰椎间盘突出症诱导的神经根性疼痛。
AIM:To explore the role of traditional Chinese medicine puerarin in radicular pain induced by lumbar disc herniation(LDH).METHODS:Rat model of lumbar disc herniation was induced by autologous nucleus pulposus(NP)implantation,and puerarin was successively intraperitonea injection for 7 d(100 mg·kg^(-1)·d^(-1)). Pain thresholds were assessed by mechanical paw withdrawal threshold(PWT)and thermal paw withdrawal latency(PWL).Spinal protein levels of Toll like receptor 4(TLR4)and phosphorylation of nuclear factor-κB(p-NF-κB,p-p65)were determined by Western blot. Spinal microglia activation was detected by immunofluorescence. Spinal expression of inflammatory and anti-inflammatory cytokines was evaluated by ELISA.RESULTS:Puerarin alleviated allodynia and hyperalgesia induced by NP implantation. Puerarin reduced the protein levels of TLR4 and p-p65,and inhibited activation of spinal microglia. Puerarin decreased the levels of inflammatory cytokines interleukin-1β(IL-1β),IL-6 and tumor necrosis factor-α(TNF-α),and increased the levels of anti-inflammatory cytokine IL-10 in myeloid tissue.CONCLUSION:Puerarin alleviates radicular pain from LDH by inhibiting TLR4/NF-κB signaling pathway-mediated inflammatory responses of microglia.
