东北地区主栽杨树品种的SSR分析及鉴定 被引量:6

SSR Analysis and Identification of Main Poplar Varieties in Northeast China

摘要 以东北地区杨树新品种、良种以及广泛栽培的已知品种为研究对象,对各无性系进行SSR分析,从分子水平对40个无性系进行指纹图谱构建与遗传关系分析,利用各无性系的指纹图谱制成包含DNA遗传背景、引物信息、扩增产物片段长度等信息丰富且易于获取的二维码,为杨树品种鉴别、知识产权保护以及苗木产业化提供技术支撑。结果表明:利用18对引物对40个杨树品种进行扩增,共扩增出101个条带,多态性比率达100%,平均每个引物扩增出5.61个条带。18对引物的产物条带的片段长度处于34~393 bp之间,引物PMGC2030扩增的条带数量最多,引物PN1297275、PMGC2885扩增出的条带数量均最少。构建了40个品种的DNA指纹图谱,开展了各杨树品种遗传相似性分析,各品种遗传相似系数处于0.48~0.89之间,并绘制了40个杨树品种的聚类分析图。对40个杨树品系进行了二维码设计,内容包括品种名称、遗传背景、拉丁名、品种类型、品种权人、扩增片段数量和长度共7项,可以更为方便地进行信息共享与利用。 Forty known poplar varieties suitable for northeast China were used for SSR analysis, and the genetic background, the primers information and the molecular length of products were recorded into the molecular two-dimensional code, so as to provide technical support for poplar variety identification, intellectual property protection and seedling industrialization. Eighteen pairs of primers were used to amplify 40 poplar varieties, and 101 bands were amplified, with a polymorphism ratio of 100% and an average of 5.61 bands were amplified for each primer. In 18 pairs of primers, the primer PMGC2030 amplified the largest number of bands, primers PN1297275 and PMGC2885 both amplified the least number of bands, and the molecular length of the product bands was between 34 and 393 bp. The DNA fingerprints of 40 varieties were successfully constructed. The genetic similarity analysis of each poplar species was carried out. The genetic similarity coefficient of each poplar variety was between 0.48 and 0.89, and the cluster analysis tree graph of 40 poplar varieties was drawn. The QR code of 40 poplar strains was designed, including the QR code of seven items including the variety name, genetic background, Latin name, variety type, variety right and the number and the length of amplified fragments, which can be more convenient for information sharing and utilization.
