卵巢癌患者组织标本HOXA5 IGF-I STIP1表达与疾病预后的相关性

The relationship between the expression of HOXA5,IGF-I,STIP1 and the prognosis of ovarian cancer

摘要 目的探讨卵巢癌患者组织标本同源异型盒基因5、胰岛素样生长因子-I、磷酸化应激诱导蛋白1的表达及其与预后的相关性。方法对卵巢癌组(80例卵巢癌患者)、良性组(35例卵巢良性肿瘤患者)、正常组(35例正常卵巢组织患者)的卵巢组织标本进行细胞染色,采用免疫组化链菌素亲生物素-过氧化酶连接法检测卵巢组织标本同源异型盒基因5、胰岛素样生长因子-I、磷酸化应激诱导蛋白1表达,并根据染色结果评定阳性表达状况。比较3组间以及不同病理特征、预后卵巢癌患者上述指标阳性表达率,并分析其阳性表达率与临床特征、不良预后的相关性。结果卵巢癌组同源异型盒基因5阳性表达率显著低于良性组、正常组(P<0.01),胰岛素样生长因子-I、磷酸化应激诱导蛋白1阳性表达率显著高于良性组、正常组(P<0.01)。相关分析显示,卵巢癌患者同源异型盒基因5阳性表达率与临床分期、淋巴结转移、不良预后呈显著负相关(P<0.05或0.01),与组织分化程度呈显著正相关(P<0.05);胰岛素样生长因子-I、磷酸化应激诱导蛋白1阳性表达率与卵巢癌患者临床分期、淋巴结转移、不良预后呈显著正相关(P<0.05或0.01),与组织分化程度呈显著负相关(P<0.05或0.01)。结论卵巢癌患者组织标本同源异型盒基因5、胰岛素样生长因子-I、磷酸化应激诱导蛋白1呈异常表达,三者均参与病情进展、淋巴结转移,且与不良预后密切相关。 Objective To investigate the relationship between the expression of homologous box gene 5(HOXA5),insulin-1ike growth factor-I(IGF-I),stress induced phosphoprotein 1(STIP1)and the prognosis of ovarian cancer.Methods Cell staining was performed on the ovarian tissue samples of the ovarian cancer group(80 patients with ovarian cancer),the benign group(35 patients with benign ovarian tumors),and the normal group(35 patients with normal ovarian tissue).The immunohistochemistry Streptavidin Peroxidase Con-jugated Method was used to detect the expression of HOXA5,IGF-I,and STIP1 in ovarian tissue samples.The positive expression status was evaluated according to the staining results.The positive expression rates of the above indicators in patients with ovarian cancer with different pathological characteristics and prognosis were compared among the three groups,and the correlation between the positive expression rates and clinical characteristics and poor prognosis was analyzed.Results The positive expression rate of HOXA5 in the ovarian cancer group was significantly lower than that of the benign group and the normal group(P<0.01).The positive expression rate of IGF-I and STIP1 were significantly higher than those of the benign group and the normal group(P<0.01).Correlation analysis showed that the positive expression rate of HOXA5 in patients with ovarian cancer was significantly negatively correlated with clinical stage,lymph node metastasis,and poor prognosis(P<0.05 or 0.01),and significantly positively correlated with the degree of tissue differentiation(P<0.05).The positive expression rate of IGF-I and STIP1 was significantly positively correlated with the clinical stage,lymph node metastasis,and poor prognosis of patients with ovarian cancer(P<0.05 or 0.01),and significantly negatively correlated with the degree of tissue differentiation(P<0.05 or 0.01).Conclusions The tissue samples of patients with ovarian cancer have abnormal expressions of HOXA5,IGF-I,and STIP1,all of which are involved in disease progression a
