摘要
目的:应用不同剂型四氧嘧啶(ALX)诱导孕兔糖尿病模型,探索一种安全、稳定的妊娠期糖尿病模型的建立方法。方法:健康新西兰孕兔54只随机分为三组,均禁食12 h后,A组经耳缘静脉注射15 ml生理盐水;B组、C组分别按100 mg/kg、130 mg/kg体重经耳缘静脉注射5%的ALX,模型不成功者分别按该剂量重复给药。1周后经耳缘静脉抽血检测空腹血糖值,以空腹血>16.7 mmol/L作为造模成功的标准。比较三组模型的成功率、流产率及死亡率。结果:A组对照组模型成功率、流产率及死亡率均为零;B组模型成功28只,流产8只,其中流产后因高血糖死亡1只,成功率77.8%,流产率22.2%,死亡率2.8%;C组模型成功4只,流产8只,其中流产后因高血糖死亡4只,成功率33.3%,流产率66.7%,死亡率33.3%。各组间相比诱导成功率、流产率及死亡率均差异具有统计学意义(P<0.05)。结论:应用ALX制备孕兔糖尿病模型,是一种相对经济、安全、可靠的方法,用小剂型多次给药的方法优于一次性大剂量给药,降低了流产率、死亡率,提高了模型成功率。
Objective Induce the diabetes model in pregnant rabbits by the different dosage forms alloxan(ALX)injection,in order to explore a method for establishing the safe,steady gestational diabetes model.Method 54 healthy gestation New Zealand rabbits were randomly divided into three groups and were fasting for 12 hours before ALX injection,then group A were injected 15 ml of normal saline by ear border vein;Group B and C were injected 5%of ALX according to 100 mg/kg and 130 mg/kg body weight by ear marginal vein respectively.The failure models were infected the same dose again.After a week,the rabbit was regarded as a successful model if fasting blood-glucose>16.7 mmol/L according to detecting the blood sampling of ear border vein.Compare the rates of success,miscarriage and mortality in the group of A,B and C.Results In group A,the rates of success,miscarriage and mortality were all 0;In group B,the success were 28,the abortion were 8,only 1 died from hyperglycemia after miscarriage.The rates of success,miscarriage and mortality was 77.8%,22.2%and 2.8%respectively;In group C,the success were 4,the miscarriage were 8 and 4 rabbits died from hyperglycemia after miscarriage.The rates of success,miscarriage,mortality was 33.3%,66.7%and 33.3%respectively.The experiment results were significantly different among Group B,Group C and the control group A.The experiment results were significantly different between Group A and B,A and C,B and C(P<0.05).Conclusion Establishing the diabetes model of pregnant rabbit by ALX is an economy,safe and reliable method relatively.Multiple low doses are better than only large dose in reducing the rates of abortion,mortality and improving the rate of success.
