新型整体柱固相微萃取与液相色谱/质谱联用分析植物中痕量细胞分裂素 被引量:1

Analysis of trace cytokinins in plants by novel monolithic column solid-phase microextraction-liquid chromatography-tandem mass spectrometry

摘要 建立了新型整体柱固相微萃取-超高效液相色谱/串联质谱分析植物中10种痕量细胞分裂素的分析方法。通过1,4-二苯基丁二炔与六(3-巯基丙酸)二季戊四醇酯巯-炔点击聚合制备了通透性较好的固相微萃取整体柱,采用红外光谱、扫描电镜进行了表征。研究了最佳萃取条件和该方法对细胞分裂素的富集能力,结果表明,方法线性范围为0.0250~5.00μg/L,检出限在2.42~8.02 ng/L之间,相对标准偏差(RSD)小于6.2%。该方法应用于分析水稻叶和番茄根中细胞分裂素含量,加标回收率为82.2%~116.4%,RSDs为2.5%~7.9%。方法能满足植物中痕量细胞分裂素的分析检测。 A novel method was developed for the simultaneous analysis of 10 cytokinins in plants by monolithic column solid-phase microextraction(SPME) coupled with ultra-high performance liquid chromatography-tandem mass spectrometry.The polymer monolithic columns with better permeability was prepared by mercapto-acetylene click polymerization of 1,4-diphenylbutylene with dipentaerythritol hexakis(3-mercaptopropionate).The columns were characterized by infrared spectroscopy and scanning electron microscope.Extraction conditions and enrichment efficiency were investigated.The method showed a linear range of 0.0250-5.00 μg/L and a limit of detection of 2.42-8.02 ng/L,with the relative standard deviation(RSD) of less than 6.2%.The developed method was successfully applied for the determination of cytokinins in paddy leave and pomato root,with the recoveries of 82.2%-116.4% and the RSDs of 2.5%-7.9%,and can be used for trace determination of cytokinins in plant samples.
