摘要
目的探讨非远处转移性胃癌预后相关的miRNA,并寻找其靶基因mRNA。方法通过TCGA数据库下载胃癌患者的临床信息和miRNA表达信息,筛选出影响患者术后总体生存率的miRNA,然后使用多因素Cox回归分析方法筛选出独立预后的miRNA。利用miRNA数据库查找独立预后miRNA的靶基因mRNA,并构建miRNA-mRNA相互作用的网络图,最后查找影响患者术后总体生存率的靶基因mRNA。结果筛选出13个具有统计学意义的影响患者术后总体生存率的miRNA(P<0.05),多因素Cox回归分析后发现3个miRNA(hsa-miR-652-5p、hsa-miR-379-3p、hsa-miR-4521)是影响胃癌预后的独立危险因素(P<0.05)。通过miRNA数据库查找到28个靶基因mRNA,对其进行生存分析,寻找到2个对患者术后总体生存率有统计学意义影响的mRNA(ECT2、IGF2)。结论高表达miRNA(hsa-miR-652-5p、hsa-miR-4521)提高非远处转移性胃癌患者术后总体生存率。高表达miRNA(hsa-miR-379-3p)降低非远处转移性胃癌患者术后总体生存率。
Objective To explore the prognosis-related miRNA of non-distant metastatic gastric cancer and to search for its target gene mRNA.Methods The clinical information and miRNA expression information of patients with gastric cancer were downloaded from TCGA database.The miRNAs that affect the overall survival rate of gastric cancer patients were screened by R software,and then the independent prognostic miRNAs were screened by multivariate Cox regression analysis.By using the miRNA databases to find the target gene mRNAs of the independent prognostic miRNAs,and construct a miRNA-mRNA interaction network.Finally,the target gene mRNAs which affect the overall survival rate of patients after operation were found.Results 13 miRNAs affecting the overall survival rate of patients after operation were screened(P<0.05).Multivariate Cox regression analysis showed that three miRNA(hsa-miR-652-5p,hsa-miR-379-3p,hsa-miR-4521)were independent risk factors for the prognosis of gastric cancer(P<0.05).Finally,28 target gene mRNAs were found by miRNA databases.Through survival analysis,two mRNAs(ECT2,IGF2)which had significant effected on the overall survival rate of the patients were found.Conclusion Overexpression of miRNA(hsa-miR-652-5p,hsa-miR-4521)can improve the overall survival rate of patients with non-distant metastatic gastric cancer.Overexpression of miRNA(hsa-miR-379-3p)reduces the overall survival rate of patients with non-distant metastatic gastric cancer.
