芒果苷、山柰酚和栀子苷对高尿酸血症小鼠的作用 被引量:4

摘要 目的探讨芒果苷、山柰酚和栀子苷对酵母联合腺嘌呤诱导的高尿酸血症小鼠的治疗作用。方法采用酵母联合腺嘌呤诱导小鼠高尿酸血症模型,测定给药后对小鼠胸腺、脾脏和肾脏指数、血清中尿酸(UA)、肌酐(Cr)和尿素氮(BUN)的含量及黄嘌呤氧化酶(XOD)活性的影响,HE染色观察肾脏组织病理学变化。结果与模型组比较,芒果苷、山柰酚和栀子苷各剂量组均能降低高尿酸血症小鼠血清中UA的含量,并能改善肾脏组织的病理损伤程度;芒果苷高剂量组和山柰酚低、中、高剂量组及栀子苷中、高剂量组均可降低血清XOD活性。结论芒果苷、山柰酚和栀子苷各剂量组均能通过抑制血清XOD活性降低高尿酸血症小鼠UA的含量,且较别嘌醇毒性和不良反应小,更具安全性,其中山柰酚的降UA作用最明显。 Objective To investigate the effect of mangiferin,kaempferol and gardenin on hyperuricemia mice induced by yeast combined with adenine.Methods The hyperuricmia model of mice was induced by yeast extracts and adenine.The effects of administration on thymus index,spleen and renal index,the contents of uric acid(UA),creatinine(Cr)and blood urea nitro(BUN),and the activity of xanthine oxidase(XOD)in serum were determined and the pathological changes of kidney issue were observed by HE staining.Results Compared with the model group,all the dose groups of mangiferin,kaeniphenol and gardenin could reduce the contents of UA in serum of hyperuricemia mice and improve the degree of pathological damage of kidney tissue.Mangiferin high dose group,kaempferol low,middle and high dose group,and gardenin middle and high dose group could reduce the activity of XOD.Conclusion All the dose groups of mangiferin,kaempferol and gardenin could reduce serum UA in mice with high uric acid by inhibiting the activity of serum XOD,and the toxicity and side effects were less and the safety was better,among which kaempferol had the most obvious therapeutic effect.
