摘要
为提高软枣猕猴桃优良种质资源的种苗繁育速度,本试验以软枣猕猴桃野生雌株资源2851的叶片为外植体,研究植物生长调节剂及MS和1/2MS对不定芽诱导、增殖、生根的影响。结果表明:软枣猕猴桃叶片的不定芽诱导培养基为MS+3 mg·L^(-1) TDZ+0.5 mg·L^(-1)6-BA+0.2 mg·L^(-1)2,4-D+7 g·L^(-1)琼脂+30 g·L^(-1)白糖,再生率达到67.2%;不定芽增殖培养基为MS+3 mg·L^(-1) TDZ+0.1 mg·L^(-1) NAA+7 g·L^(-1)琼脂+30 g·L^(-1)白糖或者MS+1 mg·L^(-1)6-BA+0.5 mg·L^(-1) ZT+7 g·L^(-1)琼脂+30 g·L^(-1)白糖,增殖系数分别为4.7和4.9;组培苗生根培养基为1/2 MS+0.2 mg·L^(-1) IBA+7 g·L^(-1)琼脂+30 g·L^(-1)白糖,生根率为91.6%。
In order to improve the breeding speed of fine germplasm resources of Actinidia arguta,the leaves of wild female resources of 2851 was used as the explant in this experiment,we studied the effects of plant growth regulators,MS and 1/2 MS on adventitious bud induction,adventitious bud proliferation and rooting of tissue culture seedling.The results showed that the Actinidia arguta of adventitious bud of leaves regeneration medium was MS+3 mg·L^(-1) TDZ+0.5 mg·L^(-1)6-BA+0.2 mg·L^(-1)2,4-D+7 g·L^(-1) agar+30 g·L^(-1) sugar,the regeneration rate could reach 67.2%;adventitious bud proliferation culture medium was MS+3 mg·L^(-1) TDZ+0.1 mg·L^(-1) NAA+7 g·L^(-1) agar+30 g·L^(-1) sugar or MS+1 mg·L^(-1)6-BA+0.5 mg·L^(-1) ZT+7 g·L^(-1) agar+30 g·L^(-1) sugar,the proliferation coefficients were 4.7 and 4.9,respectively;and rooting culture medium was 1/2 MS+0.2 mg·L^(-1) IBA+7 g·L^(-1) agar+30 g·L^(-1) sugar,the rooting rate was 91.6%.
