摘要 双向糖转运蛋白SWEET(Sugars will eventually be exported transporters)基因在植物生长发育、生理代谢及植物—病原菌互作等方面具有重要的调控作用。为了探索甘蔗SWEET基因在生长发育中的表达情况及对逆境胁迫的响应,本研究以甘蔗‘新台糖22号’为试验材料,克隆了一个甘蔗SWEET基因,命名为ShSWEET2a,并对其蛋白理化性质进行生物信息学分析和表达模式研究。序列分析表明ShSWEET2a包含一个840 bp开放阅读框,编码279个氨基酸。利用qRT-PCR技术对不同植物器官和不同非生物胁迫类型下ShSWEET2a基因表达进行分析,结果表明ShSWEET2a基因在甘蔗根、茎、叶中均有表达,茎、叶中表达量最高;在PEG、NaCl和ABA胁迫处理下,ShSWEET2a表达量均有上调,在SA胁迫处理下,ShSWEET2a表达量受到抑制。研究结果将为进一步研究SWEET基因在甘蔗生长发育过程中的作用提供理论基础。 Sugars will eventually be exported transporters(SWEET)gene has important regulatory effects on growth,physiological metabolism and plant-pathogen interaction in vivo.In order to explore the expression of SWEET gene in sugarcane growth and development and its response to stress,we cloned a sugarcane SWEET gene named ShSWEET2a using sugarcane cultivar“ROC”22 as experimental material,and carried out bioinformatics analysis on the physicochemical properties of the protein and its expression pattern.Sequence analysis showed that ShSWEET2a contained an 840 bp open reading frame encoding 279 amino acids.Quantitative PCR(qRT-PCR)was used to analyze the expression of ShSWEET2a gene in different plant organs and under different abiotic stress types.The results showed that ShSWEET2a gene was expressed in roots,stalks and leaves of sugarcane,with the highest expression in leaves and stalks.Under PEG,NaCl and ABA stress,the expression of ShSWEET2a gene was up-regulated,meanwhile,ShSWEET2a expression was inhibited under SA stress.The results of this study will provide a theoretical basis for further research on the role of SWEET gene in sugarcane growth and development.
