摘要
核桃(Juglans regia L.)是新疆的重要经济树种。为研究‘新温81’(W81)、‘新温179’(W179)和‘新萃丰’(X10)3个新疆核桃品种差异的遗传基础,我们对3种核桃仁的转录组进行了测序和分析。结果表明,所有样本与参考基因组比对后匹配率在93%以上,共有36812个基因至少在六大数据库(NR,SwissProt,Pfam,COG,GO和KEGG)中一个数据库获得注释,占总基因数目的84.97%。分组比对显示,W81与X10、W81与W179、W179与X10间分别有1921、2058和1253个差异表达基因。通过GO功能注释显示,3个品种差异基因在生物过程、细胞组分和分子功能3大类别均有分布,且分类结果相似。KEGG代谢通路富集分析表明,W81与W179,W81与X10间的差异基因主要富集在氨基酸代谢、脂肪酸合成与降解等代谢途径,W179与X10间仅在四条代谢途径有差异基因富集。本研究为培育优质核桃品种提供了科学依据。
Walnut(Juglans regia L.)is an important economic tree species in Xinjiang.In order to explore the genetic basis of differences among three Xinjiang walnut varieties,'Xinwen81'(W81),'Xinwen179'(W179)and'Xincuifeng'(X10),the transcriptome sequencing and analysis of their kernals were carried out.The results showed that mapped reads of all samples were more than 93%in comparison with the reference genome,and a total of 36812 genes were annotated in at least one of six databases(NR,SwissProt,Pfam,COG,GO and KEGG),accounting for 84.97%in all genes.There were 1921,2058 and 1253 differentially expressed genes(DEGs)among W81 and X10,W81 and W179,and X10,respectively.It was found that DEGs among three walnut varieties were distributed in three categories:biological process,cell composition and molecular function by GO functional annotation.Furthermore,the classification results were similar.KEGG metabolic pathway enrichment analysis indicated that DEGs between W81 and W179,and DEGs between W81 and X10 were mainly enriched in the metabolic pathways such as amino acid metabolism,fatty acid synthesis and degradation.However,DEGs between W81 and W179 were enriched in only four metabolic pathways.The results of this study provide scientific evidence for cultivating walnut verities with high-quality.
