海南莺歌海盐场土壤可培养微生物的多样性分析 被引量:1

Analysis of the Diversity of Culturable Microorganisms of Soil in Yinggehai Saltern,Hainan

摘要 莺歌海盐场是中国华南地区重要的食用盐生产基地。本研究从莺歌海盐场土壤中分离到可培养细菌及真菌,采用16S rDNA、ITS序列构建系统发育树,分析菌株分类学地位。结果表明,10株细菌菌株分为3个类群,菌株YGHYCB1、YGHYCB2、YGHYCB4、YGHYCB6、YGHYCB9与Bacillus acidicola、B.shackletonii及B.pseudomycoides亲缘关系较近;菌株YGHYCB1与B.amyloliquefaciens、B.velezensis聚类;菌株YGHYCB3、YGHYCB7、YGHYCB8、YGHYCB10与BreviBacillus limnophilus聚类。12株真菌菌株分为4个类群,菌株YGHYCF1与Phragmotaenium spp.聚类;菌株YGHYCF2、YGHYCF4、YGHYCF7、YGHYCF12与Sporobolomyces spp.聚类;菌株YGHYCF5、YGHYCF6、YGHYCF9、YGHYCF10与Dermatosorus cyperi聚类;菌株YGHYCF3、YGHYCF8、YGHYCF11与Moesziomyces、Pseudozyma、Oryza、Macalpinomyces 4个不同属聚为一大支,展现了这3株菌株在分类学地位上的差异。以上结果反映了莺歌海盐场土壤中可培养真菌资源多样性丰富,而细菌由于生长受到高盐环境抑制,多样性不显著。 Yinggehai Saltern is an important salt production base in South China.In this study,culturable bacteria and fungi were isolated from the soil of Yinggehai saltern.Phylogenetic tree was constructed by 16 S rDNA and ITS sequences,and the taxonomy of the isolates were analyzed.The results show that 10 bacterial isolates were divided into 3 groups.The isolates YGHYCB1,YGHYCB2,YGHYCB4,YGHYCB6 and YGHYCB9 were closely related to Bacillus cacidicola,B.shackletouii and B.pseudomycoides;YGEHYCB1 clustered with B.amyloliquefacieus and B.velezensis;YGHYCB3,YGHYCB7,YGHYCB8,YGHYCB10 clustered with BreviBacillus limuophilus.Twelve fungal isolates were divided into 4 groups.YGHYCF1 clustered with Phragmotaeuium spp.;YGHYCF2,YGHYCF4,YGHYCF7,YGHYCF12 clustered with Sporobolomyces spp.;YGHYCF5,YGHYCF6,YGHYCF9,YGHYCF10 clustered with Dermatosorus cyperi;YGHYCF3,YGHYCF8,YGHYCF11 and Moesziomyces,Pseudozyma,Oryza,Macalpinomyces were clustered into one clade,which showed the difference of taxonomy among 3 isolates.The above results indicated the culturable fungi resources in the soil of Yinggehai Saltern are quite diverse,while the bacteria growth is inhibited by the high-salt environment,and the diversity is not significant.
