摘要
目的:观察补肾活血法对系统性红斑狼疮(SLE)合并动脉粥样硬化模型小鼠免疫功能和主动脉斑块的影响。方法:8周龄大雌性载脂蛋白E基因敲除小鼠(ApoE^(-/-)mice)45只,给予一次性腹腔注射降植烷(pristane)0.5ml,继续喂养16周后,小鼠随机分为中药治疗组、西药治疗组和模型组3组各15只,以同周龄C57BL/6小鼠作为正常对照组。结果:与正常对照组比较,模型组尿素氮、肌酐、胆固醇和甘油三酯、免疫球蛋白IgG水平明显升高,补体C3水平下降,脾脏质量和蛋白尿增加,主动脉有较大斑块形成;中药治疗组和西药治疗组均能改善肾功能、蛋白尿、脾肿大,2组比较差异无统计学意义;中药治疗组在降低胆固醇和甘油三酯水平、IgG水平方面效果优于西药治疗组;中药治疗组与西药治疗组更能显著改善主动脉粥样硬化斑块病变范围。结论:与传统西药比较,补肾活血法对SLE合并动脉粥样硬化模型鼠的降血脂作用,改善免疫球蛋白水平和动脉斑块方面作用更明显。
Objective:To observe the effect of kidney-tonifying and blood-activating method on immune function and aortic plaque in mice with systemic lupus erythematosus and atherosclerosis.Methods:Forty-five eight-week-old female apolipoprotein E-gene knockout mice were intraperitoneally injected with 0.5ml norphytane.After 16 weeks of feeding,the mice were randomly divided into three groups,the Chinese medicine treatment group,the Western medicine treatment group and the Model group,15 mice in each group.C57BL/6 mice of the same age were used as normal Control group.Results:Compared with the Control group,the levels of urea nitrogen,creatinine,cholesterol and triglyceride,immunoglobulin IgG in the model group were significantly increased.The level of complement C3 was decreased,the spleen weight and proteinuria were increased,and the aorta had larger plaque formation.Both the Chinese medicine treatment group and the Western medicine treatment group could improve renal function,albuminuria and splenomegaly,and there was no significant difference between the two groups.The Chinese medicine treatment group was better than the Western medicine treatment group in lowering cholesterol,triglyceride levels and IgG levels.Compared with the Western medicine treatment group,the Chinese medicine treatment group could significantly improve the scope of aortic atherosclerotic plaques.Conclusions:Compared with the Western medicine therapy,kidney-tonifying and blood-activating therapy has more obvious effects on lowering blood lipid,improving the level of immunoglobulin and arterial plaque in SLE with atherosclerosis model mice.
