摘要
结合新型纳米传感膜材料,以黄曲霉毒素B1(AFB1)单克隆抗体为生物识别元件,通过考察AFB1与抗体之间的相互作用对测试底液中[Fe(CN)_(6)]^(3-)/[Fe(CN)_(6)]^(4-)([Fe(CN)_(6)]^(3-/4-))氧化还原体系的影响,构建了一种可用于中药材中AFB1快速检测的电化学生物传感器。在最佳条件下,对AFB1浓度的线性响应范围为0.1~100 pmol/L,检出限为14.7 fmol/L。应用该方法测定大枣、薏苡仁、决明子中AFB1的含量,加标回收率在95.3%~109.5%之间。
An electrochemical biosensor with strong specificity,high sensitivity and wide linear range was established to detect aflatoxin Bl(AFB1)in Chinese herbal medicine.In this method,the anti-AFB1 monoclonal antibodies was employed as a recognition element,and the novel nano-sensing membrane material was used to immobilize antibodies.Meanwhile,the effect of the interaction between AFB1 and antibody on the[Fe(CN)_(6)]^(3-)/[Fe(CN)_(6)]^(4-)([Fe(CN)_(6)]^(3-/4-))redox system was studied by differential pulse voltammetry(DPV)to detect AFB1.Under optimal conditions,a linear response of AFB1 concentration was obtained over the range from 0.1 to 100 pmol/L with detection limit of 14.7 fmol/L.The proposed method was used to analyse samples of Chinese herbal medicine,the obtained values of the recovery were in the range of 95.3%-109.5%.
