人参二醇型皂苷抑制肝癌细胞转移体内外实验研究 被引量:3

摘要 目的:探讨人参二醇型皂苷(PDS)体内体外对肝癌细胞转移的影响。方法:采用CCK-8法测定PDS对人肝癌Bel-7402细胞增殖的影响。利用人工基质胶模拟细胞外基质观察其对Bel-7402细胞黏附作用的影响,利用Transwell小室分析其对Bel-7402细胞迁移和侵袭的影响,通过小管形成实验观察其对人脐静脉内皮细胞(HUVEC)成管能力的影响。取昆明小鼠12只,通过尾静脉接种H22肝癌细胞悬液建立肺转移模型,将小鼠随机分为两组,分别为模型组和PDS给药组(1.4 mg/mL),每组6只。药物干预21 d,处死小鼠,取肺组织,统计肺转移结节数,称重肺脏,HE染色观察肺组织病理变化。结果:PDS能明显抑制体外肝癌细胞的黏附(P<0.05,P<0.01)、迁移侵袭和血管生成(P<0.01);体内减少肺转移结节数目,减轻肺重(P<0.01)。结论:PDS在体内外实验中均能明显抑制肝癌细胞的转移。 Objective:To investigate the effect of panaxadiol saponin(PDS)on liver cancer cells metastasis in vivo and in vitro.Methods:The effect of PDS on the proliferation of human liver cancer Bel-7402 cells was measured by Cell Counting Kit-8(CCK-8)method,and the artificial matrigel was used to simulate the extracellular matrix to observe its effect on the adhesion of Bel-7402 cells.The Transwell chamber was used to analyze its effect on the migration and invasion of Bel-7402 cells.The tubule formation experiment was used to observe its effect on the ability of human umbilical vein endothelial cells(HUVEC)to form a tube.Twelve Kunming mice were inoculated with H22 liver cancer cells suspension through the tail vein to establish lung metastasis models.The mice were randomly divided into two groups,the model group and the PDS administration group(14 mg/mL),6 mice in each group.After 21 days of drug intervention,the mice were sacrificed,lung tissues were taken,the number of lung metastatic nodules was counted,lung weight was weighed,and pathological changes of lung tissues were observed by HE staining.Results:PDS can significantly inhibit the adhesion of liver cancer cells(P<0.05 or P<0.01),migrati,invasion and angiogenesis in vitro(P<0.01),reduce the number of lung metastatic nodules in vivo,and reduce lung weight(P<0.01).Conclusion:PDS can significantly inhibit liver cancer cell metastasis in vivo and in vitro experiments.
