摘要
目的:采用网络药理学方法对苏叶黄连汤治疗慢性肾衰竭(CRF)的物质基础和作用机制进行探析。方法:运用网络数据库对苏叶黄连汤有效成分及其靶点和CRF疾病靶点进行筛选搜集。利用Cytoscape 3.7.2软件,构建中药成分-靶点网络。利用网络数据库构建苏叶黄连汤治疗CRF的蛋白质相互作用(PPI)网络,并拓扑分析出核心靶点。最后对“药物-疾病”交集靶点进行富集分析。结果:苏叶黄连汤包含26个有效成分,对应靶点484个;核心靶点涉及表皮生长因子受体(EGFR)、白细胞介素-6(IL-6)、肿瘤坏死因子(TNF)、转录因子AP-1(JUN)、72 kDAIV型胶原酶(MMP2)、雄激素受体(AR);主要生物学过程及通路有:血管平滑肌细胞增殖的正调控、溶血磷脂通路、糖尿病并发症中的糖基化终末产物及受体(AGE-RAGE)信号通路等,共7条通路,其中聚集在血管平滑肌细胞增殖正调控通路的靶点最多。结论:苏叶黄连汤治疗CRF的机制复杂,可能主要通过对血管平滑肌细胞增殖的正调控来实现治疗目的,也提示该通路是未来研究苏叶黄连汤治疗CRF的重要方向。
Objective:To explore the material basis and mechanism of Suye Huanglian Decoction(苏叶黄连汤)in treating chronic renal failure(CRF)based on network pharmacology method.Methods:The effective components and targets of Suye Huanglian Decoction and CRF disease targets were screened and collected by using network database.Using Cytoscape 3.7.2 software,the composition-target network of traditional Chinese medicine was constructed.Using the network database,the protein-protein interaction network(PPI)of Suye Huanglian Decoction in treating CRF was constructed,and the core targets were found by topological analysis.Finally,the“drug-disease”intersection targets were enriched and analyzed.Results:Suye Huanglian Decoction contains 26 effective components,corresponding to 484 targets.The core targets involved epidermal growth factor receptor(EGFR),interleukin-6(IL-6),tumor necrosis factor(TNF),transcription factor AP-1(JUN),72 kDAIV collagenase(MMP2)and androgen receptor(AR).The main biological processes and 7 pathways included:positive regulation of vascular smooth muscle cell proliferation,lysophospholipid pathway,advanced glycation end products and receptor(AGE-RAGE)signaling pathway in diabetic complications,etc..Among them,the most targets are concentrated in the positive regulation of vascular smooth muscle cell proliferation.Conclusions:The mechanism of Suye Huanglian Decoction in the treatment of CRF is complex,which may mainly achieve the therapeutic purpose through the positive regulation of vascular smooth muscle cell proliferation.It also suggests that this pathway is an important direction to study Suye Huanglian Decoction in the treatment of CRF in the future.
