摘要
目的探讨线叶菊黄酮滴丸抗耐青霉素金黄色葡萄球菌感染的作用。方法利用标准试管倍比稀释法进行体外抑菌试验;用腹腔注射细菌悬液制作感染动物模型,以动物的存活率为指标,观察其体内抗菌活性;用药物对小鼠腹腔巨噬细胞吞噬指数和淋巴细胞增殖反应的调节,进行免疫试验。结果体外抑菌试验显示线叶菊黄酮滴丸对耐青霉素金黄色葡萄球菌的最低抑菌浓度为6.25 mg·mL^(-1),对金黄色葡萄球菌的最低抑菌浓度为12.5 mg·mL^(-1)。体内抗菌试验表明,线叶菊黄酮滴丸中剂量组对感染耐青霉素金黄色葡萄球菌小鼠的死亡保护率和高剂量组对感染金黄色葡萄球菌小鼠的死亡保护率均为70%;线叶菊黄酮滴丸还可促进淋巴细胞的转化和增强小鼠腹腔巨噬细胞吞噬功能,与模型组比较,差异显著(P<0.05)。急性毒性实验未见小鼠死亡,测得其MTD值为16 g·kg-1。结论该药毒性极低,体内、外对耐青霉素金黄色葡萄球菌均有良好的抑制作用;同时可促进淋巴细胞的转化和增强小鼠腹腔巨噬细胞吞噬功能,提高小鼠的免疫功能。
Objective To investigate the role of the Filifolium sibiricum flavonoids dropping pills(FSFDP)against antipenicillin-resistant Staphylococcus aureus infection.Methods The in vitro bacteriostatic test was carried out by standard tube dilution method;The infected animal model was prepared by intraperitoneal injection of the bacterial suspension,and the animal survival rate was measured.In vivo antimicrobial activity was observed using the survival rate of the animals as an indicator;Immune testing was performed with drugs that modulate the phagocytic index and lymphocyte proliferation response of mouse peritoneal macrophages;the acute toxicity of the product was tested.Results In vitro bacteriostatic test showed that the minimum bacteriostatic concentration of geranium flavone dropping pills on penicillin-resistant staphylococcus aureus was 6.25 mg·mL^(-1),and the minimum bacteriostatic concentration on staphylococcus aureus was 12.5 mg·mL^(-1).In vivo antibacterial test showed that the death protection rate of the mice in the middle dose of FSFDF against the penicillin-resistant Staphylococcus aureus mice and the death protection rate of the high-dose group against the infected S.aureus mice were 70%;FSFDF can also promote the transformation of lymphocytes and enhance the phagocytosis of mouse peritoneal macrophages.Compared with the model group,the difference was significant(P<0.05).No acute death was observed in the acute toxicity test,and the MTD value was 16 g·kg-1.Conclusion The experimental results show that the drug has very low toxicity,and it has a good inhibitory effect on penicillin-resistant Staphylococcus aureus in vitro and in vivo.It can also promote the transformation of lymphocytes and enhance the phagocytosis of mouse peritoneal macrophages and improve the immune function of mice.
