摘要
【目的】探讨外源NO对低温胁迫下伏毛铁棒锤种子萌发及幼苗生理特性的影响,为提高伏毛铁棒锤幼苗抗寒性提供理论依据。【方法】以伏毛铁棒锤种子和幼苗为材料,采用不同浓度(0(CK),50,100,150,200,250,300μmol/L)的外源NO供体SNP溶液浸种,分别在15(对照),8,4℃下培养萌发,统计各处理的萌发率、萌发势和活力指数。在15℃下萌发伏毛铁棒锤幼苗,叶片喷施不同浓度(50,100,150,200,250,300μmol/L)的SNP溶液,以喷施蒸馏水为对照,于4℃下进行低温胁迫,通过测定超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性,以及丙二醛(MDA)、过氧化氢(H 2O 2)、可溶性糖、游离脯氨酸及叶绿素(a+b)含量,分析SNP对伏毛铁棒锤幼苗抗寒性的影响。【结果】与15℃处理相比,8和4℃低温处理下伏毛铁棒锤种子的萌发率、萌发势及活力指数均显著降低;施加SNP溶液后,随着SNP溶液浓度的提高,2个低温胁迫处理(8和4℃)伏毛铁棒锤种子的萌发率、萌发势及活力指数均呈先升高后降低的趋势,且均在200μmol/L SNP时达到最大值。4℃低温处理下,与对照相比,200μmol/L SNP溶液处理伏毛铁棒锤幼苗体内SOD、POD、CAT活性,可溶性糖、游离脯氨酸及叶绿素(a+b)含量均显著提高,MDA、H 2O 2含量均显著降低(P<0.05)。【结论】施加200μmol/L SNP溶液可缓解低温下伏毛铁棒锤种子及幼苗受到的冷害,提高种子及幼苗的抗寒能力。
【Objective】This study explored the effect of adding exogenous NO on seed germination and seedling physiological characteristics of Aconitum flavum Hand.-Mazz under low temperature stress to provide basis for improving cold resistance of Aconitum flavum Hand.-Maz seedlings.【Method】Seeds of Aconitum flavum Hand.-Mazz were soaked with different concentrations of exogenous NO donor SNP solutions(0,50,100,150,200,250 and 300μmol/L),germination was conducted at 15(control),8 and 4℃,and germination rate,germination potential and vitality index were counted.Seedlings were germinated at 15℃and leaves were sprayed with different concentrations of SNP solutions using distilled water as control.Then,seedlings were placed at 4℃for low temperature stress and activities of superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT)as well as contents of malondialdehyde(MDA),hydrogen peroxide(H 2O 2),soluble sugar,free proline and chlorophyll(a+b)were measured to analyze effect of SNP on cold resistance of seedlings.【Result】Compared with the treatment of 15℃,germination rate,germination potential and vigor index of seeds in treatments of 8 and 4℃were significantly reduced.As solution concentration increased,germination rate,germination potential and vigor index of seeds in two treatments of low temperature increased first followed by decrease with maximum values at 200μmol/L SNP.At low temperature treatment of 4℃,treatment of 200μmol/L SNP solution increased activities of SOD,POD and CAT,significantly increased contents of soluble sugar,free proline and chlorophyll(a+b),while significantly decreased contents of MDA and H 2O 2(P<0.05).【Conclusion】The application of 200μmol/L SNP solution can alleviate cold damage to seeds and seedlings of low-lying Aconitum flavum Hand.-Mazz at low temperature and improve cold resistance of seeds and seedlings.
