摘要
目的研究肾衰泄浊汤及其拆方对单侧输尿管梗阻大鼠模型(UUO)大鼠VEGF/VEGFR信号通路的影响。方法144只健康雌性SD大鼠建立UUO模型,再随机分为假手术组、补虚方组、袪邪方组、肾衰泄浊汤组、贝那普利组及模型组,每组24只,各组大鼠于第7、14、21天处死。取肾脏组织行HE、Masson染色,并在光镜、电镜下观察,评分;采用Western blot检测各组大鼠肾组织中α-SMA、VEGFA、VEGFR2蛋白表达水平。结果同时期各组比较,模型组BUN、SCr较其他组升高明显(P<0.05);治疗组BUN、SCr较其他组明显下降(P<0.05);其中肾衰泄浊汤组疗效最明显(P<0.05)。HE、Masson染色结果:除假手术组外,术后7 d各组均可观察到炎症和坏死病变,术后14、21 d可观察到炎症和纤维化,模型组纤维化程度最重。Western blot结果:假手术组α-SMA、VEGFA、VEGFR2少量表达,模型组中α-SMA、VEGFA、VEGFR2的表达随时间增长逐渐增多(P<0.05)。同时期各治疗组α-SMA、VEGFA、VEGFR2蛋白的表达较模型组均有所减少,而补虚方、祛邪方组VEGFA蛋白表达较之稍有提升(P<0.05)。肾衰泄浊汤组第21天蛋白减少较其他治疗组更明显(P<0.05)。结论肾衰泄浊汤及其拆方可能通过抑制VEGF信号通路,减少α-SMA、VEGFA、VEGFR2表达,干预UUO大鼠周细胞-肌成纤维细胞转分化过程,达到抗肾间质纤维化的作用。
Objective To study the effect of Shenshuai Xiezhuo Decoction and its decomposed components on VEGF/VEGFR signaling pathway in the rat model of unilateral ureteral obstruction(UUO).Methods A total of 144 healthy female SD rats were established UUO model,then randomly divided into sham operation group,Buxu Prescription group,Quxie Prescription group,Shenshuai Xiezhuo Decoction group,benazepril group and model group,24 rats in each group.Rats in each group were sacrificed on the 7 th,14 th,and 21 d.The kidney tissues were taken for HE and Masson staining,observed and scored under light and electron microscopes.Western blot was used to detect the protein expression levels ofα-SMA,VEGFA,VEGFR2 in the kidney tissues of rats in each group.Results At the same time of each group,the model group’s BUN,SCr increased significantly compared with other groups(P<0.05);the treatment group BUN,SCr decreased compared with other groups(P<0.05);Among them,Shenshuai Xiezhuo Decoction group has the most obvious curative effect(P<0.05).HE and Masson staining results:Except for the sham operation group,inflammation and necrotic lesions can be observed in each group at 7 days after surgery,and inflammation and fibrosis can be observed at 14 and 21 days after surgery.The model group has the heaviest degree of fibrosis.Western blot results showed that a small amount ofα-SMA,VEGFA,VEGFR2 expression in the sham operation group,and the expression ofα-SMA,VEGFA,VEGFR2 in the model group gradually increased with time(P<0.05).At the same time,the expression ofα-SMA,VEGFA,and VEGFR2 protein in the treatment group were all decreased compared with the model group(P<0.05),but the expression of VEGFA protein was slightly increased in Buxu Prescription group and Quxie Prescription group.The protein reduction of Shenshuai Xiezhuo Decoction group was more obvious than that in other treatment groups on the 21 st day(P<0.05).Conclusion Shenshuai Xiezhuo Decoction and its decomposed components may reduce the expression ofα-SMA,VEGFA,and VEGFR2 by in
