肾衰泄浊汤对肾间质纤维化小鼠HGF、TGFβ_1的调节作用 被引量:11

摘要 目的:观察中药复方肾衰泄浊汤对肾间质纤维化小鼠肾组织肝细胞生长因子(HGF)、转化生长因子β1(TGFβ1)蛋白表达的调节作用,探讨其抗肾间质纤维化的机制。方法:将60只小鼠随机分为假手术组,模型组,中药低、高剂量组,苯那普利组,采用单侧输尿管梗阻(UUO)动物模型,术后7天观察梗阻肾组织病理改变,用免疫组化方法检测肾组织HGF、TGFβ1蛋白,Ⅰ、Ⅲ、Ⅳ型胶原及FN的表达。结果:中药高剂量组和苯那普利组较模型组小鼠肾组织HGF蛋白表达增强,TGFβ1蛋白表达减弱,而Ⅰ、Ⅲ、Ⅳ型胶原、FN生成减少,两组无显著差异,而HGF表达与TGFβ1表达呈明显负相关。结论:肾衰泄浊汤可能是通过诱导HGF蛋白表达,抑制TGFβ1表达,抑制Ⅰ、Ⅲ、Ⅳ型胶原及FN等ECM的过度沉积,加强肾保护作用,对抗肾间质纤维化。 Objective: To investigate the effects of shenshuai xiezhuo decoction on hepatocyte growth factor (HGF), transforming growth factorβ1(TGFβ1) in renal interstitial fibrosis mouse. Methods: 60 mice were randomly divided into five groups: sham operation group, model group, shenshuai xiezhuo decoction high dose group, low dose group and benazepril group. The model were made by UUO, the renalhistopathological change were evaluated, the expression of HGF, TGFβ1, collagen I, III,IV and FN in renal tissue were examined by immunohistochemistry. Results: The expression of HGF in high dose group and benazepril group was significant higher than that in control group, TGFβ1, collagen I, III, IV and FN were lower, but no obvious statistical differences between the two groups. The expression of HGF was negatively correlated with the expression of TGFβ1. Conclusion: Shenshuai xiezhuo decoction can up-regulate the expression of HGF, inhibit the TGFβ1 expression, inhibit the proliferation and accumulation of ECM in renal interstice,strengthen its renotrophic function and retard the development of renal interstitial fibrosis.
