摘要
为了研究北五味子、黄芪、麦冬复合多糖和人参皂苷(SAOG)对环磷酰胺(CTX)所致免疫抑制小鼠的免疫调节作用,并阐述其作用机制。将ICR小鼠分为对照组、模型组、SAOG低、中、高剂量组[0.4、0.8、1.6 mg/(10 g·bw)],连续灌胃21 d。在给药第17天,模型组和SAOG各剂量组腹腔注射CTX 20 mg/(kg·bw),对照组腹腔注射同体积的生理盐水,连续5 d。给药结束后,测定各组小鼠肝脏指数,血清溶血素水平并对肝脏组织进行形态学观察。检测各组小鼠血清中IL-1β含量。流式细胞术检测鼠外周血中CD3^+、CD4^+、CD8^+T淋巴细胞亚群。结果显示:与对照组比较,模型组小鼠肝脏损伤明显。与模型组相比,SAOG组小鼠肝脏指数、血清溶血素水平、IL-1β水平、CD4^+/CD8^+T细胞百分比显著升高(P<0.05),肝脏的病理损伤明显减轻。表明SAOG可缓解CTX对小鼠的免疫抑制作用。
In order to study the immunomodulatory effects and mechanism of Schisandra,Astragalus,Radix Ophiopogonis polysaccharides and Ginsenosides(SAOG)on cyclophosphamide(CTX)-induced immunosuppresive mice.ICR mice were divided into control,model,SAOG-low,medium and high[0.4,0.8,1.6 mg/(10 g·bw)]dose groups.The mice in the SAOG groups were intragastrically administered SAOG once a day for 21 days and those from the control and model groups were administered the same volume of normal saline.Subsequently,the mice in the model and SAOG groups were intraperitoneally injected with CTX 20 mg/(kg·bw)once a day for 5 days.The mouse liver indexes were determined.The livers were histomorphologically observed.The level of serum hemolysin in mice of all the groups were observed and then the indices were compared.The content of IL-1β in the serum of each group was detected.The flow cytometry was used to detect the activity of CD3^+,CD4^+,CD8^+T lymphocyte subsets in the mice.The results indicated that compared with the control group,livers of the model group were significantly damaged.Compared with the model group,the liver index,the expression level of IL-1β,the level of serum hemolysin,the ratio of CD4^+/CD8^+T lymphocyte of the SAOG groups were significantly increased.The pathological damage of the liver was significantly alleviated.These results showed that SAOG can alleviate CTX-induced immunosuppressive effects in mice.
