摘要
目的:观察电针刺激足三里对重度失血性休克大鼠肝脏氧自由基的影响,并探讨其可能的机制。方法:取60只SD大鼠随机分为对照组、休克组(单纯休克组、休克复合穴位电针组)、复苏组(乳酸林格组、乳酸林格复合电针1组、乳酸林格复合电针2组),采用改良wigger's法复制重度失血性休克模型。休克复合电针组及乳酸林格复合电针2组,在模型制备成功即刻电针刺激双侧足三里30min,乳酸林格组及乳酸林格电针1组在复苏同时,穴位电刺激双侧足三里30min。检测肝组织NF-κB、ET-1、iNOS表达,SOD活性及MDA含量,电镜及光镜下观察肝组织病理学变化。结果:与各休克组比较,各复苏组NF-κB、ET-1、iNOS表达及MDA含量增加,SOD活性降低,肝组织病理损伤明显(P<0.05);与乳酸林格组比较,乳酸林格复合穴位电针1组及乳酸林格复合穴位电针2组以上损伤减轻,其中乳酸林格复合穴位电针2组以上指标降低显著,肝组织病理损伤较轻(P<0.05)。结论:休克期电针刺激双侧足三里穴能抑制液体复苏后ET-1及iNOS的合成,减少NF-κB活化及氧自由基的产生,减轻肝脏再灌注损伤。
Objective:To investigate the effects of electroacupuncture at“Zusanli”point on liver oxygen free radicals in the rats with severe hemorrhagic shock.Methods:Sixty SD rats were randomly divided into control group,shock groups(simple shock group,shock combined acupoint electroacupuncture group),resuscitation groups(lactate Ringer group,lactate Ringer combined electroacupuncture group 1,lactate Ringer combined electroacupuncture group 2).The hemorrhagic shock model was made through improved Wigger's method.“Zusanli”points were electro acupunctured for 30 minutes immediately after the shock models were reproduced in shock combined acupoint electroacupuncture group and lactate Ringer combined electroacupuncture group 2.The“Zusanli”point electro acupunctured for 30min at the beginning of resuscitation in lactate Ringer group and lactate Ringer combined electroacupuncture group 1.The expression of NF-κB,ET-1,and iNOS,SOD activity and MDA content of liver tissue were detected.The pathological changes of liver were observed under electron microscope and light microscope.Results:Compared with shock groups,the expressions of iNOS,ET-1,NF-κB and MDA content in each resuscitation group significantly increased,while SOD activity significantly decreased,and the pathological injury of liver tissue was obvious(P<0.05).Compared with lactate Ringer group,above injury were alleviated in lactate Ringer combined electroacupuncture group 1 and lactate Ringer combined electroacupuncture group 2,and above indicators reduced significantly in the lactate Ringer combined electroacupuncture group 2,with milder pathological changes(P<0.05).Conclusion:Electroacupuncture stimulation of bilateral Zusanli point during shock can inhibit the synthesis of ET-1 and iNOS after fluid resuscitation,reduce the activation of NF-κB and the production of oxygen free radicals,and alleviate the injury of liver reperfusion.
