维生素K2(MK-7)生产菌株纳豆芽孢杆菌的诱变选育及其工艺优化 被引量:4

摘要 维生素K2(menaquinone-7,MK-7)是一种人体必不可少的维生素,具有抗凝血、预防骨质疏松及抗肝癌等功能。全反式天然维生素K2(MK-7)的合成主要是由微生物发酵法而得。为了获得高产MK-7且传代稳定的生产菌株,以1株实验室前期分离自纳豆生产MK-7的纳豆芽孢杆菌Bacillus natto ND-1为出发菌株,采用常压室温等离子体(ARTP)诱变技术进行菌株诱变。经过1羟基2萘甲酸(HNA)抗结构类似物的初筛,发酵后比较MK-7浓度来复筛,获得了具有高效生产MK-7且传代稳定的突变株ND-1-A27,MK-7产量达到48.05 mg/L,是原始菌株的1.60倍。在此基础上,对突变株ND-1-A27发酵培养基组分及其培养条件进行了优化,最适发酵工艺为甘油70 g/L、大豆蛋白胨180 g/L、酵母粉30 g/L、K2HPO40.06 g/L、种子液接种量2%、发酵温度40℃、静置培养。在最佳工艺条件下,突变菌株ND-1-A27生产MK-7的产量达到(53.88±2.03)mg/L。 Vitamin K2(menaquinone⁃7,MK⁃7)is an essential vitamin for the human body.It has the functions of anticoagulation,prevention of osteoporosis,and anti⁃hepatocarcinoma.The synthesis of all⁃trans natural vitamin K2(MK⁃7)is mainly obtained by microbial fermentation.To obtain strains with the characteristics of fine hereditary stability and high⁃yield production of MK⁃7,Bacillus natto ND⁃1 isolated from natto in our lab was mutated by atmospheric and room temperature plasma(ARTP)technology.One strain designated as ND⁃1⁃A27 was obtained.The concentration of MK⁃7 was 48.05 mg/L,almost 1.60 times that of the original strain.Furthermore,the fermentation medium and the culture conditions were optimized.Under the optimized conditions with glycerol 70 g/L,soy peptone 180 g/L,yeast powder 30 g/L,K2HPO40.06 g/L,seed solution inoculum 2%,fermentation temperature 40℃,static culture,the final concentration of MK⁃7 reached(53.88±2.03)mg/L.
