摘要
通过筛选柚苷酶产生菌3-54菌株孢子的敏感抗生素最低致死浓度,采用NTG对出发菌株孢子进行诱变处理,将诱变后的孢子涂在含纳他霉素致死浓度(2.0 U/mL)的平板筛选培养基上,获得了大量的抗药性致死突变株。再结合透明圈法,经过透明圈初筛和多次摇瓶复筛,从180个纳他霉素抗性基因突变株中筛选得到1株摇瓶发酵单位达770.06 U/mL的3-54-NTG-16号菌株,比出发菌株的产酶能力提高了近100%。
After testing the resistance of sensitive antibiotic to naringinase - producing strains NO. 3 - 54, a lot of chemical resistant mutants were screened after the spores regenerated on the lethal media when they were treated with NTG. Through the transparent circle screening and sever rotation -flask experiments, the high- yielding strain 3 -54 -NTG- 16 was obtained from the 180 mutants,the productivity of which is 770.06U/mL, almost 100% higher than the original strain's in the rotation- flask experiments.
