摘要
目的:构建肥胖大鼠的模型,探讨有氧运动对肥胖大鼠血清及海马中微小RNA-221-5p(miR-211-5p)的影响,并关注有氧运动对下游因子ATP结合盒转运子A1(ABCA1)、S-100B蛋白(S-100B)和脑源性神经营养因子(BDNF)的影响。方法:使用5周龄清洁雄性大鼠36只,随机分为3组,正常对照组(C组)、高脂膳食组(HF组)和高脂膳食+有氧运动组(HE组),每组12只。C组应用普通饲料喂养,HF和HE组均应用高脂饲料喂养,均喂养10周。HE组进行10周60min/d的无负重有氧运动,每周6d。实验结束后应用实时荧光定量PCR(qPCR)检测各组血清和海马组织中miR-211-5p的含量,应用Western Blot检测各组海马中ABCA1和S100B的表达,应用免疫组化方法检测各组海马组织中BDNF的表达。结果:三组血清和海马组织中miR-211-5p的表达差别均有统计学意义(P<0.05),血清和海马组织中miR-211-5p的表达具有正相关性(P<0.05)。HF组中ABCA1和BDNF的表达明显均明显低于C组,S-100B的表达明显高于C组;HE组中ABCA1和BDNF的表达均明显高于HF组,S-100B的表达明显低于HF组(P<0.05)。结论:10周的高脂膳食能成功诱导出肥胖大鼠模型,血清及海马组织中miR-211-5p的表达均明显升高。有氧运动对肥胖大鼠血清和海马组织中miR-211-5p及下游因子ABCA1、S-100B和BDNF均有调节作用。有氧运动可能通过降低miR-211-5p的表达减少神经细胞损伤因子的生成,增加神经营养因子的分泌,发挥对神经细胞的保护作用。
Objective:To construct a model of obese rats,investigate the effects of miR-211-5p of serum and hippocampus by aerobic exercise,pay attention to the effects of downstream factors of ABCA1,S-100B and BDNF.Methods:36 clean male rats were randomly divided into three groups(12 rats in each group):normal control group(group C),high-fat diet group(group HE)and high-fat diet plus aerobic exercise group(group HE).Group C was fed with common diet,while group HF and HE were fed with high fat diet for 10 weeks.The group HE underwent weightless aerobic exercise for 60 min/d for 10 weeks,6 days a week.MiR-211-5p was detected in serum and hippocampus tissue by real-time fluorescence quantitative PCR in three groups.ABCA1 and S-100B were detected by Western Blot,BDNF was detected by immunohistochemical method in hippocampus of three groups.Results:Expression of miR-211-5p was significantly different in serum and hippocampus of three groups(P<0.05).Positive correlation was found between serum and hippocampus of miR-211-5p(P<0.05).Expressions of ABCA1 and BDNF was lower in group HF than that in group C,and expression of S-100B was higher than that in C group.Expressions of ABCA1 and BDNF was higher in group HE than that in group HE,expression of S-100B was lower than that in group HF(P< 0.05).Conclusion:High fat diet for 10 weeks can successfully induce obese rat model.Expression of miR-211-5p is significantly increased in serum and hippocampus.Aerobic exercise can regulate the levels of miR-211-5p and downstream factors ABCA1,S-100B and BDNF in serum and hippocampus of obese rats.Aerobic exercise may play a neuroprotective role by reducing the expression of miR-211-5p and increasing the secretion of neurotrophic factors.
