摘要
目的:研究卵巢子宫内膜异位症(OEMs)患者血浆中miR-17-5p、miR-20a、miR-199a、miR-141的表达情况,分析这些miRNA在内异症疾病诊断及预后评估中的效能。方法:选取前期研究中手术确诊的56例OEMs患者的血浆标本,其中32例在术后20个月内复发,24例在术后30个月的随访期内未见复发。收集同期40例非内异症人群的血浆标本做正常对照。RT-qPCR法检测miR-17-5p、miR-20a、miR-199a、miR-141表达。结果:与非内异症正常人群相比,OEMs患者血浆中miR-199a表达显著升高,miR-17-5p、miR-20a及miR-141表达显著降低,差异有统计学意义(P<0.05)。OEMs复发组血浆中miR-199a表达高于未复发组,miR-17-5p及miR-20a表达低于未复发组。miRNA-20a、miRNA-17-5p及miRNA-199a的ROC曲线下面积(AUC)分别为0.83(95%CI为0.741~0.919)、0.76(95%CI为0.641~0.892)、0.71(95%CI为0.583~0.831)。结论:检测血浆中miRNA-20a、miRNA-17-5p及miRNA-199a表达水平,可指导OEMs的临床诊断及预后评估。
Objective:To detect the expressions of plasma miRNAs(miR-17-5p、miR-20a、miR-199a、miR-141)between endometriosis patients and negative controls,and to evaluate the potential of these miRNAs as diagnostic and prognostic markers for endometriosis.Methods:Fifty-six patients were selected,32 of them had recurrence within 20 months after surgery,whereas the other 24 had not recurrence at least 30 months after surgery.Plasma of 40 healthy women was also collected.Total RNA was extracted from each sample.The relative expression levels of candidate miRNAs(miR-17-5p、miR-20a、miR-199a、miR-141)were detected using RT-qPCR.Result:The relative expression level of miR-199a was up-regulated in endometriosis patient samples compared with control samples,whereas miR-17-5p,miR-20a,miR-141 were down-regulated in endometriosis patients.The relative expressions of miR-199a,miR-17-5p and miR-20a can be used to discriminate between recurrence and non-recurrence endometrioma.The diagnostic value of miR-199a,miR-17-5p and miR-20a with area under the curve of ROC was 0.83(95%CI=0.741~0.919)、0.76(95%CI=0.641~0.892)、0.71(95%CI=0.583~0.831),respectively.Conclusions:The circulating miRNAs miR-199a,miR-17-5p and miR-20a could potentially serve as noninvasive biomarkers for endometriosis.
